以攪拌增加氧氣傳送對Aspergillus niger醱酵的影響

Effect of Stirring on the Enhancement of Oxygen Transfer during the Fermentation of Aspergillus Niger

摘要


本研究以Aspergillus niger醱酵為系統,探討以不同攪拌速度對氧氣傳送及菌體生長的影響。實驗發現醱酵12小時前氧氣傳送足夠,高轉速600、500及450 rpm的醱酵菌體生長較好。在醱酵12小時後不同轉速慢慢出現氧氣傳送不足現象以600 rpm最快出現,高轉速其乾菌重曲線在醱酵24小時後便落後較低轉速的乾菌重曲線。在醱酵32小時時氧氣傳送嚴重不足,高速攪拌600 rpm及500 rpm乾菌重已減少,而450 rpm以下之醱酵其乾菌重還在增加中。在低轉速300 rpm之攪拌速率下,其菌體濃度反而比500 rpm或600 rpm來得大;所以增加攪拌速率並未能有效地增加菌體濃度。在低轉速300 rpm的醱酵中菌體形成菌球較小,而且醱酵液中有很多的游離菌絲出現。在高轉速500 rpm或600 rpm的醱酵中菌體形成菌球體積較大並且沒有游離菌絲產生。

This research was to investigate the effect on of different stirring speed on the oxygen transfer and colonial growth in the system of Fermentation of Aspergillus niger. The experiment revealed that in the first twelve hours the oxygen supply was sufficient and the fermentation colony grew well at 600, 500, and 450 rpm; in the last twelve hours, insufficient oxygen supply gradually appeared at different rpm, earliest at 600 rpm; the dry cell weight curve after 24 hour of fermentation at higher rpm was lower than the dry cell weight at lower rpm. After 32 hour of fermentation, the oxygen supply was very insufficient at higher rpm of 600 and 500, but the dry cell weight was still increasing at 450 rpm. On the low spinning speed, 300 rpm stirring speed, the concentration of colony was higher than 500 or 600 rpm on the contrary; so the increase of stirring speed could not increase the colony concentration. On the low spinning speed of fermentation, the pellet of colony was smaller but there were a lot of free hypha appearing the fermentation fluid. On the high spinning speed 500 to 600 rpm, the pellet of colony was larger but no free hypha formation.
