魚類細胞株長期培養的自發性轉形

Spontaneous Transformation of Fish Cell Lines in Long-term Culture

摘要


本實驗探討黑鯛(Acanthopagrus schlegeli)的BPS-1、BPS-4(源自脾臟)和BPK(源自腎臟)等三種細胞株在長期培養過程中的自發性轉形。在形態上:BPS-1細胞株為表皮樣細胞,BPS-4細胞株為纖維樣細胞,二者均會長成緻密單層細胞層;而BPK細胞株則包括表皮樣和纖維樣細胞二種,會由培養初期的疏鬆的單層細胞層演變成緻密的細胞層。經長期培養後,三者細胞在基質上附著能力漸低、鋪展能力漸小、有遺傳的不穩定性及低的表面附著效應(plating efficiency)。隨著培養代數的增加,BPS細胞表面附著效應有下降的趨勢;但於雙層軟瓊脂內群落的形成則有增加的趨勢;不過族群加倍時間則無變化。而BPK細胞則隨著培養時間的增加,族群加倍時間有縮短的趨勢,且無法自軟瓊脂內形成群落。

Three cell lines derived from spleen and kidney of black porgy (Acanthopagrus schlegeli), designated as BPS-1, BPS-4, and BPK, were determined their transformed changes in long-term culture. The BPS-1 cell line and BPS-4 cell line consist of epithelial-like and fibroblast-like cells, respectively and form dense, confluent mono-layers. Whereas BPK cells, contain both epithelial-like and fibroblast-like cells, and would be transformed gradually from a radomly oriented and loosely confluent mono-layers to a dense confluent ones. Cells in long-term culture, they all show a decrease in cell-substrate adhesion and less well spread, genetic instability and low plating efficiency In BPS cells, the plating efficiency decreased with the increases of passage level and formation of colony in soft agar increased with increasing time in culture BPK cells did not formed any colony and shorten the population doubling time in long-term culture.
