不同品種及產地薏苡籽實之化學指紋圖譜建立

The Chemical Fingerprints of Seed Extracts of "Coix lacryma-jobi" of Two Varieties Grown at Different Locations

摘要


薏苡具有豐富的營養價值與機能性成分,是我國傳統藥食同源食材之一,具有調節血脂及血糖、輔助抑制腫瘤、增強免疫和抗發炎及調節內分泌等功能,為了瞭解不同品種及產地來源的薏苡其萃取成分差異,進而了解國產薏苡和進口產品之間是否有所差異,本研究以超音波輔助甲醇萃取方式,比較不同產地來源、不同品種薏苡籽實在280nm之HPLC化學指紋圖譜,作為成分特徵與含量比較之依據,共篩選5個高穩定性及再現性之吸收峰,作為成分特徵。薏苡精白後各個主要吸收峰面積損失85~97%,顯示糙薏苡麩皮含有主要UV 280nm吸收成分。臺中三號和臺中選育四號的指紋圖譜接近,不同產地包括大村、大雅與二林之間的指紋圖譜和以吸收峰面積所推估的成分含量也相當接近,其中3個吸收峰具有高穩定性,可作為未來品管依據。薏苡素的含量在種殼最高,其次為麩皮,而在精白薏苡中無法檢出薏苡素。此外,本試驗所建立之薏苡籽實化學指紋圖譜,可作為未來比較不同品種與產地薏苡萃取成分比較的依據。

Job's tears (Coix lacryma-jobi) is traditional Chinese medicinal foods that is nutritious and contains rich functional metabolites. It has been proved to improve hyperlipidemia and hyperglycemia, inhibit tumor growth, enhance functions of immune system, anti-inflammation and regulate endocrine hormones. In order to understand the variation of seed extracts in different varieties and cultivation area. Ultrasonic p rocess assisted methanol extraction was applied for all samples included in this study. The extracts were analyzed by HPLC/UV at 280 nm to compare with its chemical fingerprints. Using present chromatographic condition, 5 major peaks stably representing more than 90% of the total absorption area are identified. After seeds polishing, the absorption peaks decreased by 85-97%, indicating that most of the UV absorption metabolites exist in bran. The chemical fingerprints and amounts of each metabolite are very similar between varieties Taichung No. 3 and Taichung Selection No. 4 grown at Da-Tsueng, Da-Ya and Erh-Lin. In the LC chromatogram, 3 peaks showed stable and reproducibile which can be applied for quality control in the future. The content of coixol can be detected by HPLC. With GC/MS, 5 reported coix lactams and coixol can be detected and relatively quantified. The chemical fingerprints of seed extracts can be employed as basis for the identification of different origin.
