培養基質及植物生長調節劑對草莓微體繁殖之影響

Effect of culture medium and plant growth regulators on strawberry micropropagation

摘要


本試驗以草莓‘桃園一號’走莖爲培植體,比較培養基中添加不同濃度6-benzylaminopurine (BA)對芽體增殖及不同生長素和培養基質對芽體發根之影響。結果顯示每重複平均芽體增殖數以1/4量MS(Murashige and Skoog)添加0.5或1 mg L^(-1) BA之處理最高,芽體數分別爲2.6及2.7,但添加BA較不利根之發生。芽體培養於不添加生長素之洋菜培養基或分別以珍珠石、蛭石和粗砂爲培養基質之發根率均可達90%以上,且處理間差異不顯著,每株平均根數亦以不添加生長素之洋菜、珍珠石及粗砂處理最高,分別爲6.5、6.0及5.6,添加Indole -3- butyric acid (IBA)及1- naphthylaceticacid (NAA)生長素之發根率及根數則顯著較低且芽體基部及根呈現肥厚現象。

To compare the effect of bud proliferation on different 6-ba concentration medium and effect of bud rooting under different plant growth regulators (PGR) and culture media by using strawberry 'Taoyuan No. 1' running stalk as explants in this study. Results showed that a single bud culture on 1/4MS with 0.5 or 1 mgL^(-1) BA added has the highest proliferation average bud number of 2.6 and 2.7, respectively, however, BA added was unfavorable to rooting . Bud rooting could reached above 90% when explants was culture on PGR-free medium of agar, perlite, vermiculate, and sand with non-significant difference. The average root number per shoot on no PGR medium of agar, perlite, and sand was 6.5, 6.0 and 5.6 respectively. Medium with IBA and NAA added, the rooting incidence rate and number was obviously low and the base of bud and root was hypertrophy.
