樟芝發酵液凍乾粉之30天短期慢性動物毒性評估

A 30-day Subacute Toxicity Assessment of "Antrodia cinnamomea" Cultured Mycelium in Chinese Kun Ming Mice

摘要


本試驗對樟芝發酵全液凍乾粉(Antrodia cinnamomea freeze-dried mycelium)進行急性毒性試驗及大鼠30天重複劑量安全性評估試驗,急毒試驗結果發現對雌雄小白鼠的急性經口LD50均大於20 g/kg BW/日,以急性毒性半數致死量毒性分級,屬無毒級物質。本實驗設低、中、高三個劑量,即0.5、1.5、4.5 g/kg,另設空白對照組。每組各20隻,雌雄各半,連續餵養30天。每日進行實驗動物之臨床觀察,且每週測量受試動物之體重及攝食量。30天後取鼠血進行血液學、生化學、電解質及尿液檢查。大體解剖觀察有無明顯病變並進行組織病理學檢查。試驗結果顯示,試驗期間所有受試動物生長情況良好,無明顯臨床症狀差異發生,血液學檢查,生化學檢查,尿常規檢查,主要臟體比(organ weight ratio)及組織學檢查結果與對照組相比,亦均無明顯差異。

This study aims to assess the median lethal dose (LD_(50)) and the no observable adverse effect level (NOAEL) of Antrodia cinnamomea freezedried mycelium (Ac) in mice. Results showed that Ac has an LD_(50) above 20 g/kg body weight. In the subacute toxicity study, 40 male and 40 female mice were divided into 4 groups including a control group, namely 0.5 g/kg, 1.5 g/kg, 4.5 g/kg and the control. Animals were gavaged orally once each day for 30 days consecutively. Daily clinical observations and weekly animal body weight and feed consumption measurements were carried out. Animals were sacrificed on the 31^(st) day for hematology, biochemistry, electrolyte and urine tests. Autopsy showed no significant lesions. All tested parameters were comparable to their respective control groups with no Ac-related toxicity.
