評估口服不同乳酸菌株產品對於降低沙門氏菌侵入肉雞及其引發之發炎反應

Evaluation of the Oral Feeding Effects of Different Lactic Acid Bacteria Strains on the Salmonella Invasion and the Induced Inflammation of Chicks

摘要


利用益生菌作為動物保健之訴求之飼料添加產品功能性日益增加,因此有效的產品評估方法愈顯示其重要性。本研究以家禽動物試驗進行乳酸菌產品抑制沙門氏菌侵入及發炎,。探討雞隻對抗沙門氏菌侵入及發炎的影響。一日齡雞隻口服餵食四株乳酸菌(Lactobacillus acidophilius LA01、L. plantarum LPL01、L.fermentum LF03 和Enterococcus spp. EF03)第1天到第3天後,於第4天感染沙門氏菌。感染後第3天及第6天犧牲取其肝臟、脾臟、盲腸內容物及盲腸扁桃體並測定沙門氏菌數。另一方面使用即時定量聚合酶鏈鎖反應(real-time quantitative PCR, RT-Q PCR)測定雞隻盲腸扁桃體中促發炎細胞激素(IL-1、IL-6及IFN-)和抗發炎細胞激素(IL-10)的表現。在抑制沙門氏菌方面,所使用之乳酸菌株皆能抵抗沙門氏菌侵入約1~2 log CFU/g器官值左右。在抗發炎結果中發現口服乳酸菌之處理組與沙門氏菌感染組相比較,在感染後三天其促發炎細胞激素皆有較低的水平(p<0.05)。這些結果顯示以上述四株乳酸菌株能有效的抵抗沙門氏侵入在雞隻之臟器、盲腸的定殖及維持腸道中的免疫平衡。本研究結果可提供相關產業及檢驗之參考。

Many probiotic products, which are claimed to be able to improve protect the health of livestock animals, have been commercialized. Accordingly, the evaluation of probiotic properties of these products is important. In this study, we used chicks as experimental animals and evaluate antagonistic effects of lactic acid bacteria (LAB) against Salmonella infection. One day old chicks were fed with different LAB, i.e., Lactobacillus acidophilius LA01, L. plantarum LPL01, L. fermentum LF03 and Enterococcus spp. EF03 from day 1 to day 3. On day 4, chicks were challenged with Salmonella. The livers, spleens, cecal tonsils and cecal contents of chicks were then removed and the numbers of Salmonella cells on day 3 and 6 post-infection were determined. On the other hand, the levels of proinflammatory cytokines, i.e., interleukin IL-1β , IL-6, interferon IFN-γ and anti-inflmmatory cytokine, i.e., IL-10, in cecal tonsils were measured by reverse transcription real-time quantitative PCR (RT-Q PC R). Regarding the effects of LAB cells against Salmonella invasion of these organs, all LAB strains used in this study were able to reduce the numbers of Salmonella per gram organ by one to two orders of magnitude. For the antiinflammatory effects, chicks fed with LAB showed lower levels of proinflammatory cytokines as compared to those of the controls on day 3 post-infection (P<0.05). These results suggest that these LAB strains exert immunomodulatory effects on chicks and could be used to protect the chicks against Salmonella invasion. Such results may be used as references for livestock industries and inspection agencies.
