CMP efSwab®-Amies broth具有保存臨床嗜氧性及厭氧性病原菌的效能

CMP efSwab®-Amies Broth is Effective in Preserving both Aerobic and Anaerobic Pathogenic Bacteria

摘要


自動化檢體接種設備如InoqulA(BD-Kiestra, USA)與WASP(Walk-Away Specimen Processor, Copan, Italy)已經在大型微生物檢驗室中廣泛使用,由於此兩種機台需要使用液態檢體進行接種,因此通常使用植絨拭子(nylon-flocked swab)配合Amies輸送培養基進行檢體採集。為了因應自動接種的要求,台灣啟新生物科技股份有限公司研發一種套組,稱為CMP efSwab®-Amies broth,用於檢體的採集與輸送。本研究根據CLSI之微生物輸送系統規範M40-A2(2014年),評估CMP efSwab®-Amies broth套組對嗜氧菌包含Neisseria gonorrhoeae(ATCC 43069)與Haemophilus influenzae(ATCC 10211)以及厭氧菌包含Peptostreptococcus anaerobius(ATCC 27337)與Bacteroides fragilis (ATCC 25285)的保存效能。為了模擬檢體輸送過程,將每種菌株懸浮液調整至相當於McFarland 0.5的濃度,菌數約為1.5×10^8 CFU/mL,然後將各懸浮液分別取100μL菌液置於植絨拭子(efSwab®),再將接種的拭子插入含有Amies broth的試管中,分別在4℃和室溫(25℃)下儲存6、24和48小時。依據CLSI標準,在4℃或室溫下儲存後,菌量計數對數值的減少(log reduction)不應大於3,另外,4℃還要求上升值不可大於1。結果顯示在4℃或室溫儲存下,H. influenzae、P. anaerobius與B. fragilis均可在48小時後可被回收,而N. gonorrhoeae在24小時後可被回收,細菌計數的對數值變化均不超過3。由於CMP efSwab®-Amies broth的採檢套組且具有良好保存細菌的能力,符合CLSI M40-A2規範,因此,可完全配合自動化檢體接種儀器的接種。

Automated specimen inoculation devices, such as InoqulA (BD-Kiestra, USA) and WASP (Walk-Away Specimen Processor, Copan, Italy), have become widely used in large microbiology laboratories. As these instruments require liquid medium for inoculation, it is common to use flocked swabs coupled with Amies transport medium for specimen collection. To meet the requirement of automated inoculation, the Taiwan Creative Life Science Co., Ltd. has developed a kit, referred to as CMP efSwab®-Amies broth, for specimen collection. In this study, we evaluated its efficacy according to the CLSI Microbiological Transport System Specification M40-A2 (2014) on both aerobic bacteria including Neisseria gonorrhoeae (ATCC 43069) and Haemophilus influenzae (ATCC 10211) and anaerobic bacteria including Peptostreptococcus anaerobius (ATCC 27337) and Bacteroides fragilis (ATCC 25285). To simulate the process of specimen transport, each bacterial suspension was adjusted to McFarland 0.5 with a bacterial coun t of approximately 1.5×10^8 CFU/mL, and 100 μL each of such suspensions was placed on a flocked swab (efSwab®). The inoculated swab was then inserted into the tube containing Amies broth and stored at 4°C or room temperature (25°C) for 6, 24, and 48 hours. According to the CLSI standard, the decrease in the log value of bacterial count of a specimen should not be greater than 3 and the increase value should not be greater than 1 after being stored at 4℃ or room temperature. Results showed that H. influenzae, P. anaerobius, and B. fragilis could be recovered after 48 hours and N. gonorrhoeae could be recovered after 24 hours of storage at 4°C or room temperature with a change in the log value of bacterial count less than 3. As these results met the CLSI M40-A2 standard, we conclude that the CMP efSwab®-Amies broth specimen collection kit is compatible with automated specimen inoculation instruments.
