戊糖乳桿菌(Lactobacillus pentosus)GK4代謝物具有口腔保健功效

Evaluation of the Metabolites of Lactobacillus pentosus GK4 in Oral Health Care

摘要


各種細菌為齲齒(dental caries)、牙周炎(periodontitis)、甚至口腔癌前病變(oral potentially malignant disorders, OPMD)等之病原菌。其中,變異鏈球菌(Streptococcus mutans)、伴放線聚集桿菌(Aggregatibacter actinomycetemcomitans)、牙齦卟啉單胞菌(Porphyromonas gingivalis)等是造成齲齒(蛀牙)和牙周炎的主 要原因。在台灣,盛行率偏高的口腔黏膜下纖維化(oral submucous fibrosis, OSF)是OPMD的症狀之一。後生元(postbiotics)為益生菌(probiotics)的發酵代謝物,可能含有抗菌、抗發炎物質,並且具有高穩定性、安全性、易加工等產品化特點,適合做為口腔保健成分。本研究評估戊糖乳桿菌(Lactobacillus pentosus)GK4之發酵代謝物的口腔保健功效。體外試驗結果顯示,GK4的後生元對口腔病原菌的生長及其生物膜(biofilm)形成具有抑制作用。細胞試驗結果指出,GK4後生元製劑具有抑制口腔黏膜下纖維化纖維母細胞(OSF fibroblasts, OSFs)的遷移能力與膠體收縮能力(colloid contractility),可減少口腔相關的病變發生。這些發現,吾等認為GK4後生元具有口腔保健之功效,可被開發成為一種口腔保健產品。

Various bacteria are causative pathogens of dental caries, periodontitis, or oral potentially malignant disorders (OPMD). Among them, Streptococcus mutans, Aggregatibacter actinomycetemcomitans, and Porphyromonas gingivalis are the leading causes of tooth decay and gum disease. The high prevalence of oral submucosa fibrosis (OSF) in Taiwan is an attribute to OPMD. Postbiotics are metabolites of probiotics and may contain antibacterial and anti-inflammatory substances. In this study, we evaluated the effects of the metabolites of Lactobacillus pentosus GK4 on oral health. In vitro test results showed that the postbiotics of GK4 were inhibitory to the growth of bacteria involved in tooth decay and periodontitis and their biofilm formation. Results of cell assay indicated that GK4 postbiotics inhibited migration ability and colloid contractility of OSF fibroblasts. These findings suggest that the postbiotics of GK4 have beneficial effects on oral health and can potentially be developed to become an oral health care product.
