官能基交聯劑在膠原蛋白生物組織材料上的應用

Modification of Collagen Materials with Cross-Linking Reagents

摘要


臨床上常需要用到的生物組織材料來修補或代替病變的生物組織,本文裡介紹了幾類臨床上常使用到的生物組織材料,以及官能基交聯劑在膠原蛋白生物組織材料上的應用。基本上,官能基交聯劑可以用來交聯膠原蛋白,以增強其物理、化學以及生物的穩定性。醛類官能基交聯劑是目前臨床上使用最多的交聯劑,然而過去臨床上長期使用的經驗,發現用醛類交聯劑的膠原蛋白材料仍然遭遇到了許多問題。近年來一項過去長期使用的環氧化物交聯技術已開始被用來交聯處理各類生物人工材料器官。本文禮也分別介紹了各類醛類官能基交聯劑與環氧化物類官能基交聯劑,以及比較了它們的一些物理、化學與生物穩定特性。

collagen fibrils, reconstituted collagen fibrils, or tissues rich in collagen have been used as biomaterials or in developing artificial organs. It is known that collagen fibers constitute the fundamental framework of biological tissues. It procides a good support and framework for cell infiltration and proliferation. However, biomaterials or bioprostheses derived from collagenous tissues have to be cross-linked or chemically modified before they can be implanted in humans. The cross- linkage or chemical modification of collagenous tissues is to reduce their antigenicity and immunogenicity. Collagen can be cross-linked or chemically modified with aldehydes. However, it has been reported that the long-term performance of the biomaterials or bioprostheses cross- linked with aldehydes are suboptimal. To address this deficiency, a new cross-linking method using epoxy compounds to cross-link collagenous tissues has been developed recently. In this paper, we reviewed the biological materials available for clinical practices. Additionally, we compared the physical. chemical, and biological stability of collagen materials cross-linked with aldehydes or epoxy compounds.
