摘要


本篇是回顧在EOF的存在下,以毛細管電泳進行DNA和蛋白質線上濃縮與分離的新方法,在討論TB緩衝溶液濃度、PEO濃度和pH值及樣品間質對濃縮效率與分離的影響後,我們亦回顧利用不同PEO溶液進行步進式分離DNA之技術,另外,亦討論分析5.0μL DNA和2.1μL蛋白質樣品之線上濃縮與分離及其應用於PCR和尿液等實際樣品的分析。

We review the on-line concentration and separation of DNA and proteins in the presence of electroosmotic flow (EOF) in capillary electrophoresis (CE). The effects of concentrations of Tris-borate (TB) and poly (ethylene oxide) (PEO), pH and sample matrix on resolution and concentration factor are discussed. We also review stepwise techniques for DNA separations using different PEO solutions. Moreover, the techniques allowing on-line concentration and separation of up to 2.1-μL proteins or 5-μL DNA samples and their applications to the analysis of PCR products and urine samples have been discussed.
