利用毛細管電泳法計算主-客內包錯合物結合常數之探討

摘要


毛細管電泳法 (capillary electrophoresis, CE) 不僅在分析化學中已成為十分重要的分離工具,亦可應用於醫藥及生物科學領域。在醫藥方面,藥物分子如何辨別活性部位是十分重要的基礎,利用環糊精(cyclodextrin, CD) 與對掌型異構物之主-客 (host-guest)模型,可模擬藥物與簡單受體基質作用機制。本文將利用毛細管電泳法探討主-客分子在緩衝溶液系統中化學計量組成及內包錯合物(inclusion complexes)的遷移行為。

Capillary electrophoresis (CE) has recently achieved prominence as a very effective separation technique for analyzing large numbers of chiral and positional isomers in many research fields. The studies of molecular recognition process with artificial receptors to improve the bioavailabilities of drugs become more and more useful models in pharmaceutical industry. The mechanism of this guest- host model can be evaluated by applying the cyclodextrins (CDs) as receptors with the separation of the enantiomers by CE. In this article, the determination of binging constants by CE with measuring the change in electrophoretic mobility of a solute through buffer solution containing dissolved complexation agents is introduced, and the calculation of the stoichiometry for the inclusion complexes is also discussed.
