18^F-Labelled Radiopharmaceuticals for Positron Emission Tomography

摘要


正電子發射斷層顯像(PET)是近年來迅速發展起來的一種核醫學診斷技術,所用放射性核素氟-18(18^F)具有相對長的半衰期(t_1/2=110min),允許有較充足的藥物標記和顯像研究時間,其類氫特性不會引起標記分子的空間結構和生物活性發生明顯改變。因此18^F被廣泛用於標記葡萄糖、氨基酸、脂肪酸、核苷、受體的配體等分子作為放射性顯像劑,利用PET探查代謝、蛋白質合成和神經遞質功能活動,以診斷腫瘤、心血管、神經和精神性疾病,並指導和確定治療方案以及評價療效。標記18^F的工作和篩選適 宜的標記藥物是目前研究的重點,科研和臨床的需要在客觀上推動了18F標記藥物的迅速發展,從而對生物功能影像學和分子核醫學的發展起到了非常重要的作用。

Positron Emission Tomography (PET) is a powerful medical imaging method that has found wide applications in the last decade for research and clinical diagnosis. One of the positron emitting isotopes is fluorine-18 that is often used as a substitute for hydrogen in organic molecules with a half-life of 110 minutes, which allows complex synthesis and detailed imaging studies. Fluorine-18 has been used for the labelling of glucose, amino acids, fatty acids, nucleosides, receptor ligands and neurotransmitters for the detection of cancer, cardiovascular and neurological diseases. The study on fluorine-18 labelling and new fluorine-18 labelled radiopharmaceuticals remains to be one of the most challenging areas of PET technology in molecular imaging and nuclear medicine.
