摘要


本實驗中毛細管壁表面修飾成帶正電荷的型態的三級胺官能基之短鏈烷基胺(short chain alkyl amine)類毛細管已成功製備,除避免帶正電荷之鹼性蛋白質之吸附困擾,且具有良好的分離解析度。可於10分鐘內快速分析ppm範圍之五種鹼性蛋白質,平均分離效率達10300 plates/m,也具有良好 的再現性。背景電解質的pH值影響管壁三級胺官能基及蛋白質的質子化程度,藉由控制pH值來調整系統分離選擇性被深入探討。

A novel capillary posess short-chain tertiary amine groups was successfully prepared. The cationic bonded-phase is effective in preventing the adsorption of positively charged proteins. Fast and high-resolution separations of basic proteins in ppm range with average efficiencies of 10300 plates/m were achieved also have a good reproducibility. In addition, the protonation ratio of bonded-phase and proteins will be changed by the pH of running buffer. Thus the influence of pH were experimentally investigated in this study.
