化學計量實驗設計法結合毛細管電泳樣品線上濃縮技術於生物分析之應用

摘要


化學計量實驗設計法目的為獲得最佳化分離條件其包括部分因子實驗設計法(fFD)及中心混成實驗設計法(CCD),相較於傳統的單一變因參數探討,化學計量實驗設計法可以增加分離效率,探討參數之間的交互作用,減少實驗時間與實驗成本之損耗。毛細管電泳線上濃縮技術不需要增加儀器設備,僅利用電泳原理改變分析物本身的移動情形,配合適當的系統條件以及基礎的層析原理,便可達成濃縮的目的,反映在靈敏度上的效果可達數十倍到百萬倍,大大降低分析成本。生物檢品經適當的樣品前處理後,可利用化學計量實驗設計法結合毛細管電泳線上濃縮技術以獲得較佳的偵測感度及分離效果。

The chemometric experimental design including fractional factorial design (fFD) and central composite design (CCD) were used to optimize separation conditions. Compared to the traditional investigation, the chemometric experimental design was less time-consuming, lower in cost and can explore the interactions of parameters. The on-line sample preconcentration technology without additional equipment can increase detection sensitivity in milliontimes and lower in cost by altered the analytes' electromobility. After suitable sample pretreatment, combining the chemometric experimental design and on-line sample preconcentration technology by capillary electrophoresis can acquire the better detection sensitivity and separation efficiency in bioanalysis.
