Novel Mass Spectrometry Technology Development and Application

摘要


質譜技術是最重要的分析工具之一,質量是化合物固有的物理量,一般質譜圖只提供質量的訊息,串聯式質譜儀的二次質譜圖功能,更進一步提供化合物結構的資訊,質譜儀主要是由離子源、質量分析器、離子偵測器,真空系統及電控系統構成,由構成的要件質譜儀常面臨的問題,有游離效率、離子傳送效率、解析度、動態範圍,質量精確度,再現性、靈敏度、超大分子量生物分子測量、分析時間等,隨著基質輔助雷射脫附游離與電噴灑游離技術解決生物分子游離的問題,再加上人類基因體定序計畫的成功,蛋白質體學,醣質體學及代謝質體學興起,這些問題倍加重視,是目前質譜技術開者發努力解決的目標,本文章將報導我們關於新式游離源開發,超大分子量生物分子測量,可攜式質譜儀研發,高解析度質譜儀在蛋白質轉譯後修飾與小分子分析的工作。

Molecular weight is an inherent physical property of a molecule. Therefore mass spectrometry turned out to be one of important modern analytical tools. MS1 presented molecular weight of certain molecule. The fragmentation pattern of molecule in tandem mass spectrometry (MS/MS) used to determine structural information of a molecule. A mass spectrometer usually comprises with ionization source, mass analyzer, detector, vacuum system and data acquisition system. The mass spectrometry faced several problems such as ionization efficiency, transmission efficiency, resolution, dynamic range, mass accuracy, reproducibility, sensitivity, lager molecular detection, duty cycle, etc. After Human Genome Project (HGP) was successful to sequence complete human genome, proteomics started to set off tremendousattention. At same time, Matrix-assisted laser desorption/ionization (MALDI) and electrospray ionization (ESI) were developed to solve obstacle of bimolecular ionization. Study of proteomics coupled with mass spectrometry became important projects. In this review paper, we reported our current works related to novel ambient ionization source, large bimolecular detection, portable mass spectrometry and high resolution mass spectrometry in "Omics" and small molecules analysis.
