Adenosine 對於大白鼠攝食行為及下視丘中神經元活性之影響

Effect of Adenosine on Food Intake and Expression of cFos in the Hypothalamus

摘要


Adenosine 可有效抑制攝食行為,因此驗利用cFos 為神經元活化指標,尋找adenosine在中樞可能的作用位置。將雄性大白鼠食物剝奪2 4 小時以刺激其食慾,再以腹腔注射(intraperitoneal injection)及頸靜脈管之方式給予輸注adenosine,分別觀察攝食行為之變化和paraventricular nucleus (PVN )與supraopic nucleus(SON)中的cFos 表現情形。結果顯示100mg adenosine /kg 能明顯抑制攝食行為,並且在高劑量(I00mg/kg)顯著的增加PVN中cFos 之含量(P<0.01) ,但cFos在SON的表現情形並無顯著的差異,在給予大白鼠低劑量adenosine 10/kg 有顯著降低PVN中cFos含量(P<0.01),因此adenosine對雄性大白鼠的攝食行為的作用可能和PVN中部分神經元的活化有關。

Adenosine exerts an inhibitory effect on food intake via a central mechanism. In the present study, cFos expression was used to investigate possible sites in the hypothalamus that may be involved in the anorexigenic effect of adenosine. Male rats received intraperitoneal (i.p.) or intravenous (i.v.) injection of adenosine following 24-hr fasting to induce ingestive behavior. In the first experiment, higher dose of adenosine (l 00 mg/kg body weight, i.p.) consistently reduced food intake in fasted animals. In a subsequent experiment, cFos expression was studied in animals receiving intravenous infusion of adenosine. In the paraventricular nucleus (PVN) of the hypothalamus, there was an increase in cFos immunoreactivity in rats administered with 100 mg/kg adenosine (p<0.01), whereas a decrease in cFos was observed after given 10 mg/kg adenosine (p<0.01). However, cFos expression in the supraoptic nucleus was not different among groups. Therefore, the effect of adenosine on PVN neuronal activation was area specific, suggesting that PVN may be involved in the inhibitory effect of adenosine on food intake in fasted animals.
