M-199與NCSU-23培養液對豬卵母細胞體外成熟與後續胚發育之影響

Effects of M-199 and NCSU-23 Media on the in Vitro and Further Development of Porcine Oocytes

摘要


本試驗之目的,乃研究M-199與NCSU-23 (North Carolina University 23 Medium)培養液對豬卵母細胞體外成熟與後續胚發育之影響。卵丘-卵母細胞複合體 (cumulus-oocyte complexes, COCs) 分別培養於M-199或NCSU-23成熟培養液中48小時後,將部分去除卵丘之卵母細胞固定,檢查成熟率;其餘進行體外受精,隨後並培養於NCSU-23胚培養液中16小時與144小時,分別檢查其受精與胚發育情形。試驗結果顯示,M-199 組與 NCSU-23 組之成熟率、受精率與雄原核形成率,彼此間無顯著差異,但NCSU-23組之胚發育之比率高於M-199 組(P<0.05)。我們試驗結果證實,NCSU-23 培養液較M-199培養液有利於後續胚之發育。

The objective of this study was to investigate the effects of the M-199 and NCSU-23 media (North Carolina University 23 Medium) on the in vitro and further development of porcine oocytes. All oocyte-cumulus complexes (COCs) were cultured in M-199 or NCSU-23 medium for 48 hours, respectively. After maturation cultured, Some cumulus-free oocytes were fixed the maturation was examined the maturation rate, and the rest proceeded to fertilization in vitro. Then, the inseminated oocytes were transferred into the NCSU-23 embryo medium for 16 hours and 144 hours incubation and then fixed oocytes and examined under a phase-contrast microscope, respectively. The results showed that there was no difference between the maturation rate, fertilization rate and male pronucleus formation rate of M-199 and NCSU-23 group. However, the embryo developmental capacity of NCSU-23 group was higher than M-199 group (P<0.05). Our results demonstrated that NCSU-23 medium was beneficial for embryo development than M-199.
