本試驗之目的,乃研究PFF、HM199或HPM199對豬卵母細胞體外成熟、激活與發育之影響。豬卵丘-卵母細胞複合體分別培養於PFF、HM199或HPM199等三種不同成熟培養液中48小時後,將去除卵丘之卵母細胞置於電流強度為1.2kv/cm進行1次電激活,隨後並培養於NCSU-23培養液中16小時或144小時,俾檢查卵母細胞之體外成熟、激活與胚發育情形。試驗結果顯示,HPM199組卵母細胞之成熟率高於PFF組與HM199(P<0.05)。HPM199組卵母細胞之激活率最高(P<0.05)。然而在卵裂率方面:HPM199組高於PFF組與HM199組,且可以發育到囊胚階段。
The objective of this study was to investigate the effects of PFF, HM199 or HPM199 on the porcine oocyte in vitro maturation、activation and developmental capacity in vitro. All oocyte-cumulus complexes were cultured in PFF, HM199 or HPM199 medium for 48 hours, respectively. After maturation culture, some cumulus-free oocytes were dealing with electric activation in 1.2kv/cm field strengtht. Amended to the oocytes were subject to NCSU-23 medium for another 16 hours or 144 hours incubation and then fixed and examined. It was found that the maturated rate and activation rate in HPM199 group was the highest among three groups (P<0.05). Moreover, cleavaged oocytes only found in HPM199 treatment and some of the oocytes further developed to blastocyst.
