抗氧化及美白精油的篩選與研究

Screening and study of essential oils with anti-oxidant and tyrosinase-inhibitory activities

摘要


植物精油是萃取植物特有的芳香物質,取自於草本植物的花、葉、根、樹皮、果實、種子、樹脂等以蒸餾、壓榨方式提煉出來的。已知植物精油大約由一百多種以上的化學成分所構成,一般而言植物精油含有醇類、醛類、酸類、酚類、酮類、烯類。經由許多文獻得知許多精油的抗氧化能力很強,大多數植物精油對於2,2 -Diphenyl-1 -picryl-hydrazyl(DPPH)自由基還原能力皆有不錯的效果,但是關於植物精油美白功效之研究卻尚未有相關研究報告。首先篩選出分屬於6種科別的24種不同植物精油,比較各科植物精油對DPPH自由基還原能力,其中以樟科(Lauraceae)精油的平均還原能力41.26%最顯著,我們選定樟科植物精油中羅文莎葉(Ravaensara)與花梨木(Rosewood)精油,進一步測定其可能美白活性。細胞外酪胺酸酶活性之分析結果顯示,濃度1.5 mg/ml的羅文沙葉與花梨木精油對於酪胺酸酶酵素活性之抑制率分別為79.55 %與75.78 %,相較對照組麴酸 (Kojic acid) 濃度0.2 mg/ml抑制率為71.85 %,羅文沙葉與花梨木精油之酵素活性抑制率比麴酸高。以B16F10黑色素瘤細胞進行黑色素含量之分析,結果顯示0.112 mg/ml及0.056 mg/ml的羅文沙葉精油對於B16F10黑色素瘤細胞內黑色素含量抑制率分別為4.36%及2.18%,意味著羅文沙葉精油可能同時具備抗氧化與美白活性。

Essential oils are aromatic substances extracted from flowers, leaves, root, fruits, seeds or resins by distillation or other technology. Generally, the compositions of essential oils include alcohols, aldehydes, acids, phenols, ketones or terpenes. It is reported that essential oilsshow excellent antioxidant capacity by scavenging DPPH free radicals. However, there is no report about the skin whitening effects of any essential oil. The present study aim to screen and search potential essential oil with both anti-oxidation and whitening capacities. 24 essential oils belong to six plant families (Laminaceae, Rutaceae, Myrtaceae, Asteraceae, Lauraceae, Coniferae) were screened for DPPH scavenging activity. Among the six plant families, Lauraceae plant essential oils show the higher average DPPH scavenging activity. We choose two Lauraceae plant essential oils (Ravensara aromatica and Aniba roseodora oils) for further evaluation of potential whitening activity. The preliminary results indicated that inhibition percentage of mushroom tyrosinase of 1.5 mg/ml of R. aromatica and A. roseodora oils were 79.55% and 75.78% respectively. Furthermore, 0.056 mg/ml and 0.112 mg/ml of R. aromatica oil decreased melanin content of B_(16)F_(10) mouse melanoma cells, and the decrease percentage were 2.18% and 4.36% respectively, which indicated the potential skin care effects of R. aromatica oil in skin whitening products.
