益智仁精油之主要化學成分及其抗氧化能力分析研究

An analysis of the chemical composition and antioxidant functions of the essential oil Alpinia oxyphylla Miq

摘要


益智仁 (Alpinia oxyphylla Miq.)是具備藥理活性之亞洲地區常見天然中草藥,本研究以氣相層析質譜儀(Gas Chromatography-Mass Spectrophotometer, GC-MS)分析及鑑定其精油成分組成,並藉由體外之酚類化合物含量(Total Phenolic Contents, TPC)、硫氰酸鐵法(Ferric Thiocyanate Assay,FTC)、螯合能力(Metal Chelating Activity, MC)及β-胡蘿蔔素漂白法(β-Carotene Bleaching Assay,BCB)等四種測定方法,探討精油於不同抗氧化之效能。實驗結果得知,其前三大成分分別為Valencene (29.30%),Nootkatone (21.41%)及ρ-Cymene (10.39%);於成分類別含量中,又以倍半烯類(46.62%)含量占比為最高。抗氧化結果顯示,益智仁精油其酚類化合物含量為827.11μg ofGAE/mg,抑制過氧化物之EC50為833.31μg/mL,螯合能力EC50為813.04μg/mL,其中,以BCB試驗最為顯著,濃度在10 mg/mL時,已具73.06%抑制脂質過氧化能力,EC50為21.33μg/mL。實驗結果顯示益智仁精油具有正向之抗氧化效能。

Alpinia oxyphylla Miq. is a common natural herbs in Asia and have pharmacological activity, analysis of composition by gas chromatography-mass spectrophotometer (GC-MS) and four antioxidant abilities : the total phenolic contents (TPC) , ferric thiocyanate (FTC) , metal chelating ability (MC) and β-carotene bleaching (BCB) were studied in this paper. Three main compounds of the essential oil were Valencene (29.30%), Nootkatone (21.41%), and ρ-Cymene (10.39%). Sesquiterpenes (46.62%) is the major component of the essential oil. The antioxidant abilities are showed as follows. The TPC is 827.11 μg of GAE/mg, EC_(50) value of FTC was 833.31 μg/mL, and EC_(50) value of MC was 813.04 μg/mL. In the concentration of 10 mg/ mL the BCB value was 73.06% and EC_(50) value was 21.33 μg/mL. Among these antioxidant abilities, the BCB of the Alpinia oxyphylla Miq. Essential oil is the most effevtiveness.
