Application of Poly-aromatic Hydrocarbon Degraders for the Bioremediation of Heavy Oil Contaminated Soil

多環芳香烴降解菌應用在重油污染土壤之生物復育

Among the culture collection o f Refining and Manufacturing Research Institute o f CPC, a number o f poly-aromatic hydrocarbon (PAH) degrading strains were identified. The strains were screened and isolated from hydrocarbon-contaminated soils through either enrichment incubation or just direct isolation. Six of them degrade quinoline, benz[a]anthracene or naphthalene independently while grown in a mineral medium with Mo (molybdenum). The biodegradation ability on PAH was examined by using submerged or soil cnlture. The concentration o f PAH mixture with over 90% PAHs was 1,000μg/mL in medium. After 14 days' incubation, the whole broth or the soil was extracted with ethyl-acetate or isooctane for gradient HPLC analysis. The results showed that over 50% o f the original peaks were decreased to 20% peak area at least. Furthermore, a more significant biodegradation effect could be observed while mixed strains were applied under the same test condition. The bench scale bioremediation study was performed with a respiratory column in which the effect o f PAH degraders, biosurfactant producers, nutrients, and aeration on the degradation o f heavy oil in soils was evaluated. The results indicated that aeration was a crucial factor for a successful remediation. The heavy oil degradation rate was significantly enhanced when both PAH degraders and biosurfactant producers were inoculated simultaneously. After 56 days, the total petroleum hydrocarbon significantly decreased from 9,011 ppm to 2,168 ppm. A bio-preparation with a consortium o f PAH degraders, biosurfactant producers, extra mineral nutrients and bulking material can achieve a more efficient bioremediation in treating heavy oil contaminated soils.

台灣中油股份有限公司煉製研究所菌種庫中,有多株多環芳香烴降解菌株,係以集積培養或直接選擇方法篩選自石油污染土壤,其中六株菌種可以單獨分解奎碄(quinoline)、苯并蔥(benz[a]anthracene)或萘(naphthalene)。在液體搖瓶及土壤測試中,1000 ppm之重油混合物(含90%以上之多環芳香烴)經過約十四天後,可被這些菌株降解其中約50%的化合物達20%以上,不同菌株混合使用,能使被降解的化合物範圍更廣也更有效。簡易呼吸儀測試中,上述組合菌種配合添加適量的表面活性劑生產菌群和營養物,同時供給足夠空氣,則可更有效促進土壤中重油之降解,在56天後總石油碳氫化合物含量可由9,011 ppm降至2,168 ppm。這些多環芳香烴分解菌群和表面活性劑生產菌群所製備之微生物製劑已應用在重油污染場址,進行土壤之生物復育,同時獲致良好成效。
