自動化細胞凍管分裝系統開發與應用

Development of Automated Cell Cryovial Dispensing System

摘要


在生物醫學研究領域中,細胞的精確分裝以利後續使用是維繫細胞續存及建立安全庫存的重要關鍵。傳統細胞分裝的作業通常以人工方式進行,其步驟為將懸浮細胞液加入適量藥劑或抗凍劑,混合均勻並分裝至冷凍保存管中,並標記細胞種類、實驗日期以待後續作業。但凍管的開/關蓋與細胞液的分注為耗時費神的重複性工作,且一旦作業時程拉長,可能將使細胞數量分布不均或受到藥劑之不良影響。為有效提高作業效率與細胞存活率,本文將介紹一可自動化進行細胞凍管分裝之儀器,可達到:(1)縮短操作時程。(2)精準細胞液分注量。(3)凍管自動辨識。(4)降低細胞承受剪應力。解決目前細胞分裝作業中,人員操作造成細胞承受大量外應力、添加劑生物毒性,以及人員操作及記錄失誤、費時的問題,以大幅提高細胞存活率。

In the field of biomedical research, cryopreservation of cells is critical for maintaining cell viability and establishing safe storage. The operation of cryopreservation is usually done manually, by adding an appropriate amount of cryoprotectant to a suspension of cells, mixing them evenly, and then aliquoting them into cryovials with labels indicating cell type and experiment date for subsequent freezing. However, the repetitive steps of opening and closing cryovials and aliquoting cells are time-consuming and laborious. Additionally, prolonged processing times can lead to uneven cell distribution or negative effects on cell viability due to the cryoprotectant. To effectively increase operational efficiency and cell viability, this article introduces an automated cell cryovial dispensing instrument that can achieve: (1) shortened operation time, (2) precise cell dispensing volume, (3) automatic cryovial recognition, and (4) reduced cell shear stress. This instrument addresses the problems of toxicity caused by cryoprotectants due to manual operation and recording errors, leading to a significant increase in cell survival rates.
