應用人工授精改善台灣水牛之懷孕率

Application of artificial insemination for improving the pregnancy rate of water buffalo (Bubalus bubalis) in Taiwan

摘要


本試驗旨在了解經冷凍保存的水牛精液授精後之懷孕率,並探討水牛經同期化發情處理後,於不同時間進行人工授精之懷孕率。母水牛以PGF_(2α)兩劑間隔11天注射的方式,進行同期化發情處理,以經輸精管結紮手術後的試情公牛,進行發情偵測後,分別以新鮮或冷凍精液施以人工授精,並記錄同期化處理後,不同授精時間之懷孕率。結果顯示,新鮮精液和冷凍精液之人工授精懷孕率皆為50%(3/6和9/18);而於PGF_(2α)處理後的≦71hr、72~84hr、85~96hr及≧97hr的時距進行人工授精,其懷孕率分別為37.5%(3/8)、50%(2/4)、63.6%(7/11)及0%(0/1),並以處理後的72~96hr進行授精,可得較佳之懷孕率(50~63.6%)。另將25頭母水牛以同樣方式施以同期化發情處理,並於處理後72及96hr實施人工授精,其懷孕率為36%(9/25),其中有發情行為者,其懷孕率為45%(9/20),無發情行為者則為0%(0/5),結果顯示母水牛進行同期化發情處理後,仍需以人為或試情公牛輔助以偵測母水牛發情,以提高人工授精的懷孕率。

The objective of this experiment was to evaluate the viability of frozen semen and to study the conception rate of artificial insemination at different service timings after treatment of estrus synchronization in buffalo cows. The estrus synchronization was performed with a double dose of PGF_(2α) injection at 11- day interval. The buffalo cows were serviced twice with fresh or frozen-thawed semen after a vasectomized bull fitted with a chin-ball marker was used to detect the heat of cows. Pregnancy rates were compared after treatments of PGF_(2α). Results showed that the pregnancy rates of both fresh and frozen-thawed semen were 50% (3/6 and 9/18, respectively) and various service timings of≦72, 73 - 84, 85 - 96 and≧97 hours after PGF_(2α) injection were 37.5 (3/8), 50 (2/4), 63.6 (7/11) and 0% (0/1), respectively. It revealed that the pregnancy rate (50 - 63.6%) with service during 73 to 96 hours was highest among various service timings after PGF_(2α) injection and the frozen semen were viability for artificial insemination on water buffalo. Twenty-five Buffalo cows were given two equal dose of cloprostenol at an 11-d interval and underwent fixed-time insemination at 72 and 96 h and the pregnancy rates were 36% (9/25). Before the first service, the pregnancy rate of buffalo cows in heat and not in heat was 45% (9/20) v.s. 0% (0/5). It showed that heat detection was still important for improving the pregnancy rate after estrus synchronization.
