激勃素調控痲瘋樹株高之研究與建立轉基因系統培育高產及半矮化痲瘋樹品系

摘要


痲瘋樹 (Jatropha curcas L.) 種子含油量高達30-40%,被視為新興替代性生質能源作物。然而欲以痲瘋樹油取代現行能源尚有許多改進空間,如痲瘋樹單棵植株結果率不高,並且株高均達兩公尺以上,造成低產量及不易採收的問題,因此高產、高含油量以及半矮化品系成為痲瘋樹產業化發展的首要條件。由於痲瘋樹基因庫中缺少矮化基因,無法以傳統育種方法有效獲得符合經濟效益之雜交品系,利用分子育種方式創造痲瘋樹新品系成為唯一途徑。本論文首先以激勃素 (gibberellin) 生合成抑制劑paclobutrazol (PAC),證明降低激勃素含量確實可有效調節痲瘋樹之株高,然而人為大量噴灑藥劑的方法耗時費力,不符合產業效益。分析痲瘋樹中激勃素生合成相關基因的調控機制,比較不同器官中激勃素相關基因表現差異,結果顯示激勃素生合成基因GIBBERELLIN 20-OXIDASE 2 (JcGA20ox2) 和激勃素去活化基因GIBBERELLIN 2-OXIDASE 2 (JcGA2ox2) 可能為影響痲瘋樹株高的最主要兩個基因,並且二者對於激勃素的含量皆具有回饋機制調控 (feedback-regulatory mechanism) 的現象。最後本實驗自行建立有效的痲瘋樹再生系統與轉殖系統,藉由農桿菌媒介轉殖法 (Agrobacterium - mediated transformation),將JcGA20ox2-RNAi及35S::JcGA2ox2載體轉殖至痲瘋樹基因體中,並利用抗生素篩選建立高產及半矮化之轉基因痲瘋樹新品系。

Jatropha curcas L. is the favorable specie as alternative biofuel resource owing to the approximately 40% of oil in the seed. However, it harbors several disadvantageous characters, for example, less yield of net seeds and more than 2 meters of plant height causing inconvenient harvests and management. It is tempting to dissect functional role of gibberellins (GAs) on the growth height of J. curcas. The treatment of paclobutrazol (PAC), an inhibitor of GAs biosynthesis, brought about the dwarfism in J. curcas, indicating GAs inhibition is an available approach to obtain semi-dwarf plant. The spatial expression profile of GA-related genes revealed that GIBBERELLIN 20-OXIDASE 2 (JcGA20ox2) and GIBBERELLIN 2-OXIDASE 2 (JcGA2ox2) were two major genes playing roles in controlling plant height in J. curcas. Both of them also participated in GA-related feedback-regulatory mechanism. In this study, we established effective J. curcas regeneration and transformation system. Therefore, molecular breeding were carried out by transformation of Agrobacteriuim tumefaciens harbouring JcGA20ox2-RNAi and 35S::JcGA2ox2 in J. curcas. The results of this study would be potential to obtain high yield and semi-dwarf cultivars.
