異源四倍體LLAA“Gelria × Purple Sensation”百合組織培養再生及體外結球調控

摘要


LA型雜交百合生長呈中強簇生型,多倍體化後形態發生偏向鱗片型生長。本研究目的為探討異源四倍體選系LLAA “ Gelria × Purple sensation” 組織培養體外結球與生長發育之影響因子,期以針對較偏晚熟之特性進行改善。本試驗以百合試管苗鱗片為培植體,接種於MS基本培養基,另外參試添加有機添加物、BA、活性碳、蔗糖或ABA生合成抑制劑對其再生作用及小鱗莖生長發育之影響。結果有機添加物以1 g/L 麥芽萃取物不定芽再生率較高。在含有0.3-1.5 mg/L BA之培養基能促進鱗片葉形成。此外,培養基中添加1-2 g/L 活性碳亦有助於鱗片葉的形成。高糖濃度則有利於小鱗莖肥大生長,但會抑制葉身發育,在12%蔗糖濃度下無鱗片葉發生。鱗片葉形成數目及速率與百合的早熟性有密切的關係,8-10枚鱗片葉可視為其生理成熟的一個指標。經ELISA分析結果顯示,百合群間內生ABA含量與結球時葉原體後發育形態密切相關。早熟性百合內生ABA含量較低,呈鱗片葉型結球,而晚熟種百合內生ABA含量較高,呈鱗片型結球。而LED光源組合BRFr照射下可促進鱗片葉發生。

LA hybrid lily was mid-high vigorous rosette type in growth. Mitotic polyploidization of LA hybrid lily was scale type of growth. To improve the character of late-maturing, in this frame, we were probing the factors that influenced in vitro bulb development in LLAA “ Gelria × Purple sensation”. This work used the in vitro lilies scale as the explants cultured on MS medium supplemented with organic additive, BA, Activated charcoal (AC), sucrose or ABA inhibitors. The results showed that the optimal medium of bulb regeneration was MS+0.1 mg/L NAA+1 g/L AC+1 g/L malt extract. Modified MS medium supplemented with 0.3~0.5 mg/L BA promoted improved the scale leaf formation. The addition of 1~2 g/L AC promoted scale leaf formation. The medium supplemented with high concentration of sucrose improved size of bulblets but inhibited blade formation. On the 12% sucrose medium showed no scale leaf formation. Scale leaf number and formation rate were the physiologic mature index when 1to 8 scale leaves formation. Lilies of endogenous ABA content closely to related to the bulblet development. The distinct morphological difference between scale type and scale leaf type growth patterns in L. spesiosum and L. formosanum, respectively, we used for ELISA assays show the endogenous ABA level was significantly higher in oriental lily(L. spesiosum)than longiflorum lily(L. formosanum). This research studies on influence of regeneration and growth under different light quality shows that(BRFr)irradiation would improve the leaves formation.
