牙科用生醫材料試驗系統之建立

摘要


牙科材料在應用於人體之前,要先進行生物性評估,試驗其成份接觸人體組織時可能產生的毒性。任何新材料要使用在口腔內,都要經過整體化的測試,確定新材料所擁有的優點,遠大過任何可能產生的危險。L939 及 3T3一類穩定的老鼠纖維母細胞株常被用在牙科材料生物毒性的評估上,因為此類的細胞株生長速度穩定,用於生物實驗時的反應也具有再現性。然而,此類細胞與牙科材料所接觸到的人體細胞間本質上的差異很有可能造成實驗室內得到的結果與臨床的表現上相關性不高。由於大多數牙科材料直接接觸的是口腔內的上皮細胞,因此,如能建立一株更接近正常黏膜上皮細胞的穩定細胞株,將對於評估牙科材料有所助益。 在本篇論文中,建立了兩株本土性鱗狀上皮細胞癌之細胞株-TW1.5 及 TW2.6。TW1.5由舌部的鱗狀上皮細胞癌培養而來,TW2.6則由頰黏膜的鱗狀上皮細胞癌培養而來。TW1.5藉由培養過程中自發性的突變成為一穩定的細胞株。TW2.6則在缺乏B細胞及T細胞的小鼠體內連續數次的活體篩選而成為一穩定的細胞株。在本論文中對於此兩細胞株進行了釵h鑑定工作,包含型態的紀錄,細胞週期調節蛋白的表現,生長曲線,p53基因的定序,及染色體分析等。此兩細胞株為本土性,且角化之上皮細胞株,其特質可能更為接近正常的黏膜上皮細胞。此兩細胞株具備相當良好之生長潛力,未來相當適合用於牙科材料之測試。此兩細胞株也可應用於口腔癌之研究,探索口腔癌患者癌化的詳細機制,及用於發展新的癌症治療策略。

Biological evaluation of dental material is performed to determine the potential toxicity resulting from contact of the component materials with the body. Evaluation of any new material for oral use requires data from systematic testing to ensure that the benefits provided by the use of the materials will exceed any potential risks produced by the materials. Continuous cell lines, like L939 or 3T3 mouse fibroblasts, are being routinely used for the testing of cytotoxic properties of dental materials because their reproducible growth rates and biologic responses. Nevertheless, the question whether these are adequate target cells has often been raised with regard of the clinical relevance of data derived from in vitro studies. Most dental materials came to contact with mucosal cells, therefore, a stable cell line that is closer to normal epithelial cells is being pursuit for evaluation of these materials. In this report two native SCC (Squamous Cell Carcinoma) cell lines were developed - TW 1.5 and TW 2.6, from primary culture of squamous cell carcinoma of tongue and squamous cell carcinoma of buccal mucosa specimens, respectively. The cell line TW1.5 was immortalized through spontaneous mutation, and the cell line TW2.6 was immortalized through in vivo selection in skid mice. Characterization of these two cell lines including morphology, cell cycle regulation protein expression, growth curve, sequencing o fp53 gene, and chromosome analysis had been carried out in vitro. These two cell lines are native, keratinized, and may have characters that are similar to normal epithelium cells of mucosa. These SCC cell lines might be a good candidate for material testing because of their cellular origin and growth potential. They may also be useful for researching the possible mechanism of carcinogenesis in SCC patients and development of new cancer therapeutic strategies.
