升麻面片之化學成分研究

摘要


升麻面片為毛茛科多年生草本植物升麻的乾燥根莖,主產於中國遼寧、黑龍江、湖南及山西等地。常用於中藥材,味甘,辛,性微寒,具解熱、鎮靜等作用,內服入煎劑,外用研末調敷,煎水含嗽或淋洗。 升麻面片以甲醇浸泡兩次,浸泡液經減壓濃縮,乙酸乙酯萃取,利用管柱層析法以梯度沖提方式,以及高效能液相層析法(HPLC)做分離純化。得到的純化合物再以氫核磁共振圖譜、碳核磁共振圖譜、紅外線光譜以及質譜來推測結構。若經化學資料庫MDL Crossfire Commander 查詢為已知化合物,則與文獻值比對確認;若為尚未被發表過之新化合物,或屬天然物之首次分離,則需二維核磁共振圖譜、以及高解析質譜加以輔助,來判定其結構。 本次實驗之成果共計46 個化合物,分別為: 1.三帖類 (triterpenoids)21 個 2.香豆素類 (coumarins)5 個 3.色酮類 (chromones)4 個 4.芳香族類 (aromatic compounds)9 個 5.吲哚類(indoles)2 個 6.apocarotenoids 3 個 7.固醇類(steroids)2 個 其中10 個為新化合物,4 個為首次分離之天然物。

Cimicifuga heracleifolia (L.) distributes over mainland China. It has been used as anti-inflammatory, and antipyretic agents in Chinese medicine. The methanol extract from the root of Cimicifuga heracleifolia (L.) was concentrated to give a residue which was partitioned between ethyl acetate and water. The ethyl acetate layer was separated by column chromatography on silica gel, preparative thin layer chromatography and high performance liquid chromatography (HPLC) to give 46 compounds. Among these compounds, 10 new and 4 isolated for the first time from natural sources are elucidated. The total isolated included 21 cycloartanes, 5 coumarins, 4 chromones, 9 aromatic compounds, 2 indoles, 3 apocarotenoids, and 2 steroids. By means of IR spectra, 1H NMR spectra, 13C NMR spectra (1D and 2D techniques) and Mass spectra, Of these compounds, the chemical structures of these compounds are determined. The known compounds were identified by spectral analysis and compared with those reported in literatures or from authentic samples.
