近年來,由於混合型新興毒品在台灣極為盛行,以偽裝方式,混雜於咖啡包、奶茶包、果汁包等食品中。進而流竄於夜店、KTV、飯店派對等娛樂場所,造成年輕族群之嚴重濫用。目前已知主要成分常混合數種卡西酮類毒品,而也會搭配鎮定安眠之苯二氮平類安眠藥物,如硝甲西泮(一粒眠)或硝西泮等。因應混合型咖啡包類之新興毒品在年輕族群之氾濫,需發展相關之檢驗方法,本研究針對13種台灣近年常見新興毒品或其代謝物,如4甲基甲基卡西酮、一粒眠類緣物、新型去氯之K他命類緣物、對-甲氧基甲基安非他命、、、等等,發展液相層析串聯質譜法 (LC-MS/MS) 之尿液檢驗方法。 相對於GC-MS,LC-MS/MS方法 能減化前處理時間與層析分離的難度,因此尿液檢體以稀釋上機之方式進行。在方法確效評估結果:偵測極限皆可在0.5-2 ng/mL之間,定量極限皆可在2-5 ng/mL之間,線性範圍自定量極限到1000 ng/mL ,R2值皆大於0.995,而精密度之CV% 值與準確度皆小於10 %。最後進行實際樣品之分析評估,透過真實樣品之分析,進行相關新興毒品檢驗方法之修正與改進,並了解毒品氾濫之趨勢。
In recent years, the mixed emerging drugs prevail in Taiwan. They are mixed in coffee packets, milk tea packets or juice packets in a disguised way, scattered in entertainment venues such as nightclubs, KTV, hotel parties, etc., having resulting in serious abuse by young people. At present, it is known that the main ingredients include several mixed cathinone drugs, with the sedative and hypnotic benzodiazepine, such as nimetazepam or nitrazepam. In response to the serious abuse of mixed emerging drugs among young people, it is necessary to develop relevant testing methods. This study focuses on 13 emerging drugs or their metabolites that are common in Taiwan in recent years, such as 4-methylmethcathinone, nimetazepam analogue, new type of dechlorinated ketamine analogue, para-methoxymethylamphetamine, etc., and develop liquid chromatography tandem mass spectrometry (LC-MS/MS) urine testing. Compared with GC-MS, the method on LC-MS/MS can reduce the time of pretreatment and chromatographic separation. Therefore, the developed method is carried out by diluting urine samples and shoot them to LC-MS/MS. In the method validation results: the detection limits can be between 0.5-2 ng/mL. The quantification limit can be between 2-5 ng/mL. The linear ranges are from the quantification limit to 1000 ng/mL. The R2 values are all Greater than 0.995, while the CV% values of precision and accuracy are both less than 10%. Finally, through the analysis of real samples, the actual measurement is carried out, fulfilling the correction and improvement of the relevant emerging drug testing, and understanding the trend of drug abuse.
