鹿角靈芝對於四氯化碳誘發小白鼠肝毒性的保護作用

摘要


肝臟性疾病於台灣方面可說是極容易被發現的疾病之一,尤其近年來肝癌的發現情形更為普遍;但另一方面除了B、C型肝炎帶原者之外,由於生活環境之改變、社會、工作和家庭等壓力因素,所發生之肝臟障礙現象也非常多見,此因素或許也是導致肝癌發生機率上升之原因,對於肝臟之保健方面實驗也應該以多元化的方式來探討。 以人體生理學來看,肝臟就如一解毒工廠,負責清理所有侵入人體之異物及合成體內所需之蛋白質等功能。因此肝組織如果發生障礙時,就有可能發生中毒、肝硬化或肝癌等病症,在西醫方面雖有藥物可用來控制病情,但在於中醫學方面則可利用靈芝來保護肝臟及維護肝功能的正常化。而其所含有主要物質為三萜類化合物(terpenoid)是由6個isoprene單元組成之天然物質。靈芝含有特殊之三萜類化合物亦稱為靈芝酸(Ganoderic acid)、赤芝酸(Lucidenic acid)、赤芝酮(Lucidone)及赤靈酸(Ganodermic acid)等,根據實驗結果顯示這些化合物具有抑制癌細胞生長,抑制組織胺釋放,預防中老年人心血管方面疾病及降低血脂等生理上之效果。 本研究所採用之靈芝,俗稱鹿角靈芝(Ganoderma Amboinense或Ganoderma lucidum(antle))亦屬赤芝一族。一般靈芝之研究報告很多,但對於鹿角靈芝實際功效之實證報告除本草綱目等之記載外仍非常稀少。因此本實驗利用化學方法(即利用四氯化碳:CCl4)誘發小白鼠肝臟障礙後給予鹿角靈芝或先給予鹿角靈芝飼育再誘發肝臟障礙之方式來探討對於保肝方面之影響,以便進一步瞭解此產品對於保健方面之確實效果及提升其商業價值。

Liver diseases such as hepatitis B, C, and cirrhosis are commonly occurred in Taiwan in past 20 years, in which liver cancer is one of ten major death causes in Taiwan society.The promotive factors for the occurrence and deterioration of liver cancer include environmental variation, stress and dietary complexity.Thus, the prevention and therapy for liver diseases have been paid attention.In order to alleviate the impact from liver diseases to country, society, and individuals, the functional foods (herbs) with multiple protection for liver should be examined and developed. Liver possesses multiple physiological functions such as metabolism and detoxification.When its work is overloaded and/or too many toxic materials are consumed, hepatoxicity occurred.So far, sufficient clinical medications for liver diseases control are lack.It has been notified that Chinese herbs such as Ling-zhi may be a potent herb with liver protection.The major compounds of Ling-zhi are terpenoids, which is consist of 6 isoprone units.It has been confirmed that Ling-zhi has the following special components: ganoderic acid, lucidenic acid, lucidone and ganodermic acids.Several previous studies have demonstrated that these natural compounds could suppress cancer cell growth, inhibit histamine release, protect vascular via reducing blood lipid level. Ganoderma amboinense or Ganoderma lucidum(antle), an Ling-zhi used in this present study, contains high level of these above active compounds.Thus, the protective and therapeutic effects of this Ling-zhi will be investigated in this animal study. Mice will be treated by CCl4 to induce liver injury. The pre-intake and post-treatment of Ling-zhi will be processed.The influence of herb treatment on oxidation and inflammation will be measured.This study will provide important findings for evaluating whether this herb could be applied for liver diseases protection and therapy.
