以抗氧化分子與發光二極體對於退化性關節炎疾病之效應

Synergetic effects of antioxidant molecules and light-emitting diode (LED) on the therapy of osteoarthritis (OA) diseases

摘要


退化性關節炎(OA)是關節炎的主要形式 ; 也是最常見的退化性疾病之一。在臨床階段顯示,除了觀察此疾病引起的炎症反應。此外,許多研究已經注意到自由基(Free radical)與退化性關節炎的相互關係。 本研究利用過氧化氫誘導豬軟骨細胞產生類似於退化性關節炎(OA-like)之發炎反應,並以抗氧化分子(Antioxidant molecular)結合紅色發光二極體(Light-emitting diode)應用於此反應上的治療,且評估預防性(Pre-treat)與治療性(Post-treat)的效應是否具有去除自由基與降低發炎反應的能力。在初步結果顯示,本實驗之抗氧化分子與紅色發光二極體(635nm)具有良好之生物相容性和去除自由基及降低過氧化氫的累積,此外,發光二極體超過兩小時連續照光會抑制細胞之生物活性。最後,探討抗氧化分子結合發光二極體的效應結果顯示;在第一步抗氧化分子結果顯示,在治療發炎反應的效應比預防性的效應好。且在加入白藜蘆醇(Resveratrol)的效果比維他命E(Vitamin E)的效果好。進一步的結合發光二極體,其結果顯示;發光二極體自身能降低其IL-1β、TNF-α與增進SOX-9、CAT、COL-2的基因表現。而與抗氧化分子結合後其效應,能增進前驅發炎因子IL-1β、TNF-α向下調控(Down-regulate)與CAT、COL-2其向上調控(Up-regulate)的效果,以治療性的效應較預防性的效應好。

Osteoarthritis (OA), one of the most commonly seen degenerative disease, is a form of arthritis that caused by the breakdown and loss of cartilage of joints. In addition to focusing on the features of inflammatory reaction caused by osteoarthritis, many previous clinical observations and research results have shown that there must be a relation between free radicals and osteoarthritis. Here we used porcine chondrocytes as a model of human chondrocytes. Moreover, osteoarthritis-like inflammatory reactions was induced by hydrogen peroxide in porcine chondrocytes. The main treating strategy applied in our research was a combined treatment consisting of antioxidant molecules and light-emitting diode (LED). The pre-treated and post-treated effects of whether the free radicals were eliminated and whether the inflammatory reactions were reduced were evaluated. Previous results have shown that the antioxidants and LED (635nm) used in our research have good biocompatibility. Besides, the free radicals were effectively eliminated and the accumulation of hydrogen peroxide was apparently reduced. Finally, the effect of the combined treatment comprising antioxidant molecules and LED light were as following: In the first step of the treating procedure, antioxidant molecules were used alone, and the results showed that the post-treated effect was better than the pre-treated effect. Moreover, Resveratrol was more effective than Vitamin E. In the next step of the treating procedure, LED light exposure was added, and the outcome indicated that LED exposure alone decreased the expression of IL-1β and TNF-α gene; while the expression of SOX-9、CAT and COL-2 were increased. After combined with antioxidant, the experimental results showed that the expression of pro-inflammatory cytokines like IL-1β and TNF-α were down-regulated, and the expression of CAT and COL-2 were up-regulated. All in all, our research has shown that the combined treatment consisting of antioxidant molecules and LED light has much greater post-treated effects than pre-treated effects.
