中鏈油 (medium-chain triglyceride oil, MCT) 中所攜帶的為飽和及碳數介於6-12之間的脂肪酸鏈,由於MCT在體內的運輸途徑與氧化速率不同於長鏈油 (long-chain triglyceride oil, LCT),因此本次實驗係探討在低與高油飲食中利用MCT部分取代黃豆油對於第2型糖尿病大鼠體內脂質代謝相關酵素活性與脂質代謝的影響。實驗主要以32隻Sprague-Dawley大鼠為研究對象,以STZ誘發為第2型糖尿病後分為低黃豆油組 (Low soybean oil diet, LS)、低中鏈油組 (Low medium-chain triglyceride oil diet, LM)、高黃豆油組 (High soybean oil diet, HS) 與高中鏈油組 (High medium-chain triglyceride oil diet, HM)。實驗期8週後,分析血糖、血脂質、肝臟脂質及酵素活性,並將肝臟與生殖腺周圍白色脂肪組織進行H&E染色。結果顯示,在高油飲食中MCT的部份取代具有降低血液中LDL-C、NEFA與肝臟中TC濃度;且具有提升血液中HDL-C與HDL-C/LDL-C比值的效果。由上述結果推論MCT的給予可能藉由降低大鼠體重、調節肝臟中Acyl-CoA carboxylase、HMG-CoA reductase、Acyl-CoA oxidase與Carnitine palmitoyltransferase酵素活性達到降低血脂質與肝脂質的效果;且MCT改善效果在高油飲食中較為顯著。
MCT (medium-chain triglyceride oil), with six to twelve carbons in length, differ from other triglyceride. MCT and long-chain triglyceride oil (LCT) are absorbed and oxidize in different ways. The aim of this study was to assess replacement of soybean oil with MCT in low- or high-fat diet on lipid metabolism in STZ-induced type 2 diabetes mellitus (T2DM) rats. Thirty-two T2DM Sprague-Dawley rats were divided into low soybean oil diet (LS), low medium-chain triglyceride oil diet (LM), high soybean oil diet (HS), and high medium-chain triglyceride oil diet (HM) groups. After eight weeks, blood sugar, serum lipids, hepatic lipids and enzymes activities were measured. Results revealed that under high-fat diet, replacement of soybean oil with MCT can lower serum LDL-C, serum NEFA, and hepatic total cholesterol levels, and increase serum HDL-C concentration and HDL-C/LDL-C ratio. In conclusion, MCT reduced serum and hepatic lipids accompanied losing body weight and regulation hepatic acyl-CoA carboxylase, HMG-CoA reductase, acyl-CoA oxidase and carnitine palmitoyltransferase activities in T2DM rats. Besides, the effects are much more significant in T2DM rats fed high-fat diet.
