大豆蛋白中的 β-conglycinin 對糖尿病腎病變大鼠之影響

摘要


本研究係將 Wistar 大鼠以尾靜脈注射 streptozotocin 方式誘發糖尿病,並以含有氯化鈉的飲食和飲水加速糖尿病腎病變的進程。實驗飲食中的蛋白質以大豆蛋白和不同含量的 β-conglycinin 取代部分酪蛋白,以探討大豆蛋白和 β-conglycinin 對於糖尿病腎病變進程之影響。實驗共進行二十七週,控制組 (C group) 於實驗進行中全程餵食 AIN-93M 標準飲食,其餘各組實驗飲食皆添加 1% NaCl,蛋白質分別為:14% 酪蛋白 (D group)、7.0% 大豆蛋白 + 7.0% 酪蛋白 (P group)、1.750% β-conglycinin + 12.250% 酪蛋白 (A group) 及 3.50% β-conglycinin + 10.50% 酪蛋白 (B group)。實驗末將觀察不同飲食對實驗動物血糖、血脂、血壓及腎功能等各項指標的影響。實驗結果發現:給予大豆蛋白和 b-conglycinin 可以調節血糖和增加胰島素敏感性。在脂質代謝部份,大豆蛋白和 b-conglycinin 皆可改善血漿和肝臟中脂質生化值。且透過抑制 ACE 活性的作用、降低血壓和腎臟受損的情形,以及改善腎功能指標,顯示大豆蛋白和 b-conglycinin可以延緩此動物模式糖尿病腎病變的進展。

The objective of this study was to investigate the effects of soy protein and different dose of β-conglycinin on diabetic nephropathy in rats. In this study, rats were induced to have diabetic nephropathy with intravenous injection streptozotocin and NaCl diets. Rats were divided into the control group and four experiment groups. The duration of the experimental period was twenty-seven weeks. Diet of the experiment groups contain soy protein or β-conglycinin as substitution of part of the casein. At the end of the study, blood, liver, kidney, heart, aorta, and lung were collected for analysis. Our study demonstrated soy protein and b-conglycinin decreased blood glucose and improved insulin sensitivity. Soy protein and b-conglycinin could also regulated lipid metabolism. Otherwise, through the ACE inhibition, soy protein and b-conglycinin could decrease blood pressure and renal damage. There results suggest that dietary soy protein and b-conglycinin supplementation may have beneficial effects on delaying of the progression of diabetic nephropathy .
