荔枝,為無患子科的植物,生存在亞熱帶、中國南部和台灣等地區。荔枝的果皮呈暗紅色,有瘤狀突起,乾且薄;果肉呈白色透明狀,厚而多汁。根據文獻,荔枝籽具有良好的抗氧化活性;而具有抗氧化活性可能也會具有降低色素沉澱的效果。 將荔枝籽陰乾後,以95%的酒精萃取三次,濃縮後得598.4克。再經過分配萃取得到乙酸乙酯層98.1克、正丁醇層198.5克和水層210.8克。而經由B16-F10黑色素細胞和酪胺酸酶酵素的活性篩選,乙酸乙酯層皆有最好的效果。取乙酸乙酯層進行管柱層析。經過NMR、MS的鑑定後,共得到20個化合物。 20個化合物為10個phenolic類化合物、2個coumarin類化合物 、4個fatty acid類化合物、1個flavonoid類化合物及3個其他類化合物。其中化合物LCS8 (3,4-dihydroxybenzoic acid ethyl ester) 在抑制酪胺酸酶酵素有最好的效果。但在黑色素細胞實驗上具有細胞毒殺性。
Litchi chinensis, belonging to family Sapindaceae, is a king of subtropical fruit tree and famous for long time in Southeast Asia and Taiwan. The fruits of Litchi chinensis are called “ Lychee ” has dry, red pericarp and white edible aril. The seeds extract of lychee may reduce hyperpigmentation effect. In this study, the seeds of lychee was extracted with 95% ethanol and sequentially partitioned with ethyl acetate, n-butanol and water. Therefor, the ethyl acetate fraction with the higher inhibitory activity for tyrosinase and reduced hyperpigmentation for B16-F10 melanoma cells. The fraction was subjected to silica gel column chromatography. Twenty compounds were obtained and isolated. The structures were identified by NMR, MS and literature data. LCS 8 was inhibit tyrosinase and prevent melanin production. But it has cytotoxicity, how to reduce its toxicity with other ingredients will be the future research work.
