玉山薊對四氯化碳誘導肝損傷大鼠血清及組織中甲基乙二醛及D-乳酸的影響

The effects of Cirsium kawakamii Hayata on methylglyoxal and D-lactate in serum and tissue in CCl4-induced rat liver injury

摘要


本篇探討玉山薊(Cirsium kawakamii Hayata, CK)在四氯化碳誘導肝 毒性模型下,對於甲基乙二醛(Methylglyoxal, MG)與D-lactate 之影響,以 評估MG 及D-lactate 作為組織損傷指標之可行性。將50 隻 Sprague-Dawley(SD)大鼠隨機分為五組,除正常組外,其餘四組均每週兩 次腹腔注射四氯化碳:大豆油=4:6的混合物(1 mL/kg體重)誘導肝臟損傷, 並分別灌餵水飛薊素200 mg/kg/day、低濃度玉山薊25 mg/kg/day 或高濃 度玉山薊125 mg/kg/day,實驗為期八週。於第3、6 及8 週收集血清測定 其生化指數,並於第8 週收集肝組織,以切片病理染色鑑別肝損傷程度。 結果指出生化指數在第8 週時明顯增加,且與組織損傷量化結果一致。 在給與水飛薊素或玉山薊治療後,其血清生化指數及組織形態皆顯著改 善。此外,正常組與嚴重的肝損傷組相比其血清中MG 含量明顯地增加 (141.36 ± 7.86 vs. 245.09 ± 35.49 μM), 而肝組織中D-lactate 濃度也明顯上 升(8.56 ± 0.62 vs. 17.49 ± 2.00 nmol/mg ,正常組 vs. 損傷組),然而血清 D-lactate 在損傷組並無顯著性變化。在經過水飛薊素或玉山薊治療後, 血清MG 含量及肝臟中D-lactate 含量明顯降低(血清MG 含量 : 145.70 ± 9.32 and 119.55 ± 22.48 μM, 肝組織D-lactate 含量: 11.72 ± 1.87 and 9.64 ± 0.74 nmol/mg, 水飛薊素組及玉山薊高濃度組)。上述結果顯示,血清中 MG 含量及肝臟中D-lactate 含量極有潛力發展成為組織損傷之指標。

In this study, we investigated the effects of Cirsium kawakamii Hayata (CK) on the concentration of Methylglyoxal (MG) and D-lactate in serum and liver tissue and evaluated whether MG and D-lactate are novel markers in CCl4-induced rat liver injury. In experiment, 50 Sprague-Dawley male rats were randomly divided into 5 groups. Soybeen oil with 40% CCl4 was intraperitoneally injected into the rats to induce hepatotoxicity at volume of 1 mL/kg BW twice a week except for the normal group. Besides, 3 groups were orally administered silymarin 200 mg/kg/day, low dose CK 25 mg/kg/day and high dose CK 125 mg/kg/day for eight weeks, respectively. Sera were collected at week 3, 6 and 8, and hepatic tissues were collected at week 8. AST, ALT and histological stain were used to examine the liver injury. The results showed that AST and ALT significantly increased at week 8, and the findings were consistent with those of tissue sections as well. After silymarin and CK treatment, the blood biochemical indexes and histological morphology improved remarkably. In addition, serum MG level elevated remarkably under severe liver injuries (serum MG condition: 141.36 ± 7.86 vs. 245.09 ± 35.49 μM, liver D-lactate : 8.56 ± 0.62 vs. 17.49 ± 2.00 nmol/mg of protein, normal vs. lesion groups, respectively). However, the D-lactate level in serum shows no significant difference under damage condition. On the other hand, through silymarin and CK treatment, the MG level in serum, D-lactate level in liver were recovered obviously (serum MG : 145.70 ± 9.32 and 119.55 ± 22.48 μM, liver D-lactate: 11.72 ± 1.87 and 9.64 ± 0.74 nmol/mg, silymarin and CKH, respectively). These results indicated that MG level in serum, and liver D-lactate concentration might be used as markers of liver injury.
