近年來從草本植物中尋找良好的天然抗氧化劑,用以對抗生物體內的氧化性傷害日漸受到各界的矚目。本研究目的是評估台農一號及名間長紅兩種山藥品系之水萃物的體外和細胞氧化狀態及儲存對大汕二品對細胞氧化狀態的影響。在體外試驗測試山藥水萃取之DPPH自由基清除能力、還原力及總多酚含量;在細胞試驗乃將小鼠肝細胞(FL83B)及小鼠肝癌細胞(Hepa1-6)以氧化劑25mM t-BHP處理後,測試山藥水萃物對細胞存活率、過氧化氫及脂質過氧化物(MDA)含量的影響。結果顯示,體外試驗方面,台農一號的水萃取物具有較高的DPPH自由基清除能力;名間長紅水萃取之總多酚含量則高於台農一號;兩種山藥在還原力上則無顯著差異;細胞試驗方面,山藥水萃物雖然對FL83B 和Hepa1-6都不具有保護或修復因氧化所造成的傷害之效能,但卻顯著減少細胞內過氧化氫的含量。比較特殊的是山藥水萃物有降低FL83B細胞中脂質過氧化物MDA的含量,但在Hepa1-6細胞中,山藥水萃物反而會增加脂質過氧化物MDA的含量。因此推測山藥水萃物對細胞的抗氧化能力有其助益,值得更深入探討。
Recently, there is increasing interest in finding natural antioxidants from plants to protect human body against damage due to oxidative stress. The purposes of this study were to evaluate the antioxidant ability of water extracts from two species of Taiwanese yams, Tainung No. 1 (TNG1) (Dioscoera alata L.) and Mingjian (Dioscoera alata L. var. purpurea M. Pouch), and the effect of storage on the antioxidant ability of water extract from Darson (Dioscoera alata L.). Measurements of the antioxidative activities included total phenolic content, reducing power, a ,a-Diphenyl-b-pricryl-hydrazyl (DPPH) radical scavenging activity, and the hydrogen peroxide and malondialdehyde (MDA) contents in t-BHP treated mouse liver cell and mouse liver cancer cell. The results showed that the water extract from TNG1 had higher DPPH radical scavenging activity but less total phenolic content than the water extract from Mingjian yam. No significant difference was found in the reducing power between the two species. Although the yam extracts showed no protective effect toward both t-BHP-treated FL83B and Hepa 1-6 cells, since hydrogen peroxide content in both t-BHP-treated FL83B and Hepa 1-6 cells increased. For lipid peroxidation measurement, both water extracts from TNG1 and Mingjian yams reduced the sodium nitroprusside (SNP)-induced MDA content in FL83B cells, but not in Hepa 1-6 cells.
