本論文主要探討利用混合薄膜超過濾程序,自醱酵處理液中分離純化靈菌紅素,為了提高靈菌紅素的純度,尋找薄膜過濾之最佳操作條件。實驗採用恆壓攪拌式進行超過濾,來探討在不同操作條件之阻隔率之變化。在超過濾程序中,因受到濃度極化和結垢現象的影響,導致濾速衰減。 當進料濾液濃度增加,阻隔率上升。在YM10下,濃度為300 mg/L與500 mg/L其阻隔率在3 ~ 5 %,1000 mg/L最高可達16 %;而在YM1下,300 mg/L與500 mg/L其阻隔率在40 ~ 50 %,而1000 mg/L最高可達77 %。而在經由純度分析下,由YM10進行過濾之靈菌紅素,其濾液端純度最高可提升2倍;而YM1則是濃縮端純度較高,最高可提升2倍。最後,利用串聯薄膜程序(PES10、YM1)來提高其純度。濾速理論分析方面,利用Birgitte等人在1997年提出濾餅過濾理論(Cake filtration theory),來推得在不同操作條件下薄膜結垢指標(Modified fouling index, MFI)。
In this study, separation and purification of prodigiosin from ferment broth by ultrafiltration using regenerated cellulose(YM) and polyethersulfone(PES) membranes with MWCO 10 and 1 kDa were studied. The YM 1 kDa membrane had a higher rejection of prodigiosin than that of the 10 kDa membrane at low concentration. The rejection of prodigiosin was about 50% and 5% for 1 kDa membrane, respectively. The results for rejection at high concentration were the same. The purities of prodigiosin were purification achieved 2 fold in the permeate. Finally, the two-step membrane filtration processes was proposed. The MFI(Modified fouling index) were used to analyze flux behavior. It was shown that the flux decline was due to solute concentration polarization and fouling.
