本研究建立一個快速簡單的高效能液相層析-二極體陣列檢測器(HPLC-DAD)方法,同時偵測食品中被添加的melamine (三聚氰胺)、cyanuric acid (三聚氰酸)、ammeline (三聚氰胺二醯胺)和ammelide (三聚氰胺一醯胺)的含量。本研究使用高效能液相層析法,分離的層析管柱為Phenyl Hexyl (250 × 4.6 mm I.D.; 5 μm),以硼酸鈉緩衝溶液(100 mM,pH= 7)為液相層析之移動相,且流速控制在1.4 mL/min。以4-hydroxybenzoic acid為本實驗之內部標準品,待測物則以二極體陣列檢測器(DAD)在波長214 nm吸收偵測。偵測極限為melamine (0.05 μg/mL);cyanuric acid (0.2 μg/mL);ammeline (0.1 μg/mL) ;ammelide (0.1 μg/mL)。定量範圍分別為 melamine (0.25、2、5、20及40 μg/mL);cyanuric acid (0.5、2、5、20及40 μg/mL);ammeline (0.5、2、5、20及40 μg/mL) ;ammelide (0.5、2、5、20及40 μg/mL)。注入體積為10 ?尳,層析分析可以在4分鐘以內完成。melamine、cyanuric acid、ammeline和ammelide定量極限為0.25、0.5、0.5和0.5。 利用此方法可檢測食品中是否被添加具有腎毒性的人工添加物melamine及其衍生物cyanuric acid、ammeline和ammelide,可做為食品檢驗的參考指標。
A high performance liquid chromatographic (HPLC) method with diode array detection (DAD) is described for the simultaneous determination of melamine, cyanuric acid, ammeline and ammelide in food. The HPLC separation of the analyte was achieved on a Phenyl-Hexyl column (250 × 4.6 mm I.D.; 5 μm), using a mobile phase, consisting of 100 mM sodium borate buffer, pH 7.0 at a flow rate of 1.4 mL/min. 4-Hydroxybenzoic acid was used as internal standard. Diode array detector (DAD) was used, with melamine, cyanuric acid, ammeline and ammelide monitored at UV 214 nm. The linear range for the determination of melamine, cyanuric acid, ammeline and ammelide were 0.25 – 40 μg/mL, 0.5 – 40 μg/mL, 0.5 – 40 μg/mL and 0.5 – 40 μg/mL, respectively.
