露兜樹屬植物分布於及亞熱帶區域,在馬達加斯加及馬來西亞有露兜樹屬最多的物種數目。紅刺露兜樹(P. utilis)是馬達加斯加的原生種,在台灣為栽培種。在本次研究中,紅刺露兜樹的樹葉經由甲醇萃取,之後使用兩種不同的方式進行分配萃取,得到五個分層(乙酸乙酯層、正丁醇層、水層、二氯甲烷層以及鹼水層)。在這些分層中,乙酸乙酯層顯示有抗發炎活性,而甲醇層擁有抗氧化活性。 在一系列的純化後,25個化合物被分離出來,經由光譜分析及質譜資料的鑑定決定其結構。這些化合物包括7個lanosterol type三萜類、7個固醇類、4個cyclohexenone衍生物、2個生物鹼、2個苯類糖苷、1個木酚素糖苷。在這些化合物中,化合物1、8及9為新化合物。化合物8及9為一種新類型之化合物,具有特殊的5-(hexahydroindolizin-8-ylidene)- 3-methylfuran-2-one骨架,並且也可分類為一種新型的Pandanus生物鹼。雖然乙酸乙酯層及甲醇層分別對抗發炎及抗氧化活性測試具有效果,然而對兩類活性而言,並沒有有效之純化合物被分離出來。此一主題仍待進一步的研究。
Plants belonging to Pandanus (Pandanaceae) are native in tropical and subtropical regions, with the largest number of species in Madagascar and Malaysia. P. utilis is original in Madagascar and cultivated in Taiwan. In this study, the powder of P. utilus leaves was extracted by MeOH and partitioned in two different methods and yielded five layers (the EtOAc, n-BuOH, water, CH2Cl2 and alkalineaq layers). Among these layers, the EtOAc layer shows anti-inflammatory activity and the MeOH layer shows antioxidant activity. After a serious of purifications, twenty five compounds were isolated and the structures of the isolates were elucidated by spectroscopic methods and mass spectroscopic data. The isolated compounds include 7 lanosterol type triterpenoids, 7 steroids, 4 cyclohexenone derivates, 2 alkaloids, 2 benzyl glycosides and 1 lignan glycoside. Among them, 1, 8 and 9 are new compounds. Compounds 8 and 9 possess a novel 5-(hexahydroindolizin-8-ylidene)-3-methylfuran-2-one skeleton, which is a new chemical entity. Moreover, they also can be classified into a new type of Pandanus alkaloid. Although the EtOAc and MeOH layers show significant activities in anti-inflammatory and antioxidant assays, respectively, however, no compound was active toward two target functions, The subject needs to be further investigation.
