線球菊化學成份及生物活性之研究

摘要


菊科的線球菊屬(Grangea)植物為常見的一年生草本植物,線球菊屬植物共有14種,常見於熱帶或亞熱帶的亞洲和非洲地區。線球菊(Grangea maderaspatana)是該屬植物中唯一廣泛存在的物種。在泰國及許多國家常作為醫療用途,其葉部能用來當健胃、驅風、調經和抗脹氣用藥。 將線球菊的乙醇萃取物經一系列管柱層析純化後共得到gramaderins A-G (1‒7)等七個新化合物以及十六個( 8‒23)已知化合物。化合物的結構鑑定主要利用1D和2D NMR (COS、HMQC、 HMBC以及NOESY)等光譜數據分析以及比對文獻上已知化合物的光譜資料而確定。   將所有化合物進行抗發炎以及細胞毒殺活性篩選。5,3'-dihydroxy-3,6,7,4',5'-pentamethoxy- flavone (11)以及conyzatin (12)有抗發炎的活性數據,而所有送測的化合物則無顯著的細胞毒殺活性。

Grangea belongs to Asteraceae, and is a genus of suberect or prostate annual herb. It has fourteen species of Grangea distributed in tropical and subtropical Asia and Africa. G. maderaspatana is the only species found widely and used medicinally throughout Thailand and many countries. The leaves of G. maderaspatana are used as stomachic, carminative, an emmenagogue, and antiflatulent agents. Phytochemical investigation of the ethanolic extract of G. maderaspatana led to the isolation of seven new compounds, gramaderins A-G (1‒7), together with 16 (8‒23) known compounds. The structures were elucidated through interpretation of mass and spectroscopic methods, especially 2D NMR techniques (COSY, HMQC, HMBC, and NOESY), and compared the spectral data of related compounds. The isolates were screened for the anti-inflammatory and cytotoxic activity. Among the tested compounds, 5,3'-dihydroxy-3,6,7,4',5'-pentamethoxyflavone (11) and conyzatin (12) showed significant anti-inflammatory effect by inhibiting superoxide anion generation and elastase release from human neutrophils in response to fMLP. Moreover, all tested compounds did not show any cytotoxic effect against MDA-MB-231, HepG2, and A549 cancer cell lines.
