建立以藍胸鶉生產IgY抗體之方法
Immunization and IgY antibody production in Chinese Blue Breasted Quails
摘要
本研究旨在建立以藍胸鶉(Chinese Blue Breasted Quail)為動物模式(animal model)免疫及純化IgY之方法。優良試驗動物之禽類需要具備有生命力強、飼養容易和體型小，並可以節省飼養空間和飼糧成本，所以本土性鳥類—藍胸鶉，是具有潛力禽鳥。蛋黃免疫球蛋白(immunoglobulin yolk；IgY) 近年來已被廣泛地利用在被動免疫性治療，因為由雞蛋蛋黃中製備IgY，有許多優點，例如有雞蛋收取，不需要專業採血技術，而且雞蛋蛋黃中只有一種抗體的存在—IgY；並且禽類比牛、豬、兔子等哺乳動物較經濟，在大規模飼養上，更節省空間和花費較低，又因為由於雞蛋獲得的抗體又比由血液取得抗體衛生，從雞蛋中IgY含量比哺乳動物血清或乳汁中IgG抗體含量高，綜合上述，本研究期望以藍胸鶉免疫產出IgY試驗之建立。試驗分為三部分：第一部分為建立免疫與純化IgY之方法；第二部分為藍胸鶉對抗原免疫最低劑量之探討和IgY之功能性探討；第三部分將藍胸鶉IgY作為工具試驗，用於ELISA、Western-blot 和免疫螢光組織染色上的可行性。試驗結果指出以藍胸鶉免疫產生之IgY可以用實驗上，而且5μg goat-IgG作為最低劑量抗原，即可以使藍胸鶉被動免疫，免疫注射第3次後的第12週達到高峰。

IgY antibodies, also called egg yolk immunoglobulins, are the only immunoglobulin in egg yolk and transferred in the female from serum to egg yolk.Chinese blue-breasted quails(CBBQ) are the smallest quail in family phasianidae, and their advantages including hardiness, small body size, rapidly generation, high laying production and easily discriminating genders make them a potential avian animal model for experiments.We have established an antibody production system using this smallest quail (CBBQ). In this study we tested if we can use goat IgG as an antigen to induce antibody production in Chinese Blue-breasted Quail and wheather the IgY produced by CBBQ can be recognized by commercial anti-chicken IgYantibody.The hens were separated into several groups, three hens each, and immunized with 5μg or 150μg Goat-IgG.The eggs were collected every day. We used the Dot-blot,ELISA and western-blot to detect CBBQ-IgY immunoreactivity.We have established a first protocol of producing IgY in CBBQ.The IgY of CBBQ can be detected by commercial anti-IgY antibody.The titer and the specificity of IgY is a ideal for immunology application in the lab.
