應用放射性同位素碘131標誌抗體於肝癌治療之研究

摘要


肝癌的死亡率為國人癌症的第二名,由於肝癌血管性豐富,目前能夠利用栓塞化學栓塞治療,但是仍然有諸多限制。本研究目的是利用放射性同位素(_^131)I,標誌在單株抗體Anti-Glypican-3 (GPC3) clone 9C2上,使放射性同位素標誌之抗體在體內與肝癌細胞表面抗原進行特異性結合,分析各個器官與腫瘤之劑量,了解藥物之體內分佈與代謝途徑,並利用(_^131)I所放出之β射線,藉以達到殺死癌細胞的目的,以探討放射免疫治療在肝癌治療上的可能性。 小鼠試驗結果顯示其的確具有在體內殺死肝癌細胞的能力,且通過細胞實驗了解不同劑量下的癌細胞存活率,我們發現(_^131)I標誌單株抗體GPC3 clone 9C2具有肝癌治療上的可能性。

Liver cancer has the second mortality rates in all cancers in Taiwan. Because of well vascularity, Transarterial Chemoembolization is used for treating liver cancers nowadays. But it’s still limited by many reasons. The purpose of this study is using 131I radioisotope-labeled antibody Anti-Glypican-3 (GPC3) clone 9C2 to specifically bind to the surface antigen of hepatocellular carcinoma (HCC) in vivo, and analyzed the bio-distribution and metabolic pathways in the organ and tumor. Utilizing β-ray, radioimmunotherapy may achieve the purpose of killing cancer cells. This study reveals that the radioisotope-labeled antibody could actually kill cells of HCC in vivo. With the result of cell viability versus dose of antibody, the potential for HCC treatment by 131I labeling GPC3 clone 9C2 is promising.
