液相層析法對Memantine之製劑分析

摘要


本研究乃利用衍生化反應配合高效能液相層析-紫外光偵測法( HPLC-UV),來對臨床上用於治療阿茲海默症藥物memantine建立一簡單快速且高敏感度之微量分析方法。本研究方法使用(2-Naphthoxy)acetyl chloride (NAC)為衍生試劑,在溫和的條件下進行衍生反應。本研究對影響衍生反應之諸多變數,包括衍生試劑需要量、反應時間等均詳加探討,建立適當基本分析條件並應用於測定市售製劑中的memantine含量之分析,可測定之定量範圍是1~50μM,而偵測極限為0.03μM(注入體積10μL)。在同日間與異日間的相對標準偏差(R.S.D.)皆小於2.9%,顯示本研究法具有良好之定量可靠性,可應用於市售製劑中memantine之含量測定。

A simple,rapid and sensitive high performance liquid chromatographic method with UV detection was established for the analysis of memantine. The method is based on the derivatization of memamtine with (2-Naphthoxy) acetyl chloride (NAC) at mild conditions.This study discussed several factors of reaction, including the amounts of NAC and the reaction times, to establish the basic analysis condition for determing the quantity of commercial used memantine. This newlymethod can provide a quantitated range between 1-50 μM with detection limit on 0.03μM when injected volume is 10μL. The relative standard deviations of Intra-day or Inter-day were less than2.9%. It shows this method with good reliability of quantitatively determination for commercial used memantine.
