Hypoxylon sp. 12F0687 (炭角菌科)、Xylaria papulis (炭角菌科)、Mycoleptodiscus sp. 09F0149 (巨座殼科)及Lachnum abnorme (晶杯菌科)為台灣植物之內生真菌,分別從冬青科糊樗根部、爵床科柳葉鱗球花莖部、胡椒科薄葉風藤根部及紫金牛科雨傘仔莖部分離並培養而得。本研究共得到七個新化合物、一個天然首次分離及29個已知化合物。上述各化合物皆由各種圖譜分析決定。 於Hypoxylon sp. 12F0687中分離到三個新化合物:hypoxyphenone (1)、 hypoillexidiol (2)及(S)-(+)-vermelone (4),一個天然首次分離化合物:(S)-(–)-3-hydroxy-3-methyloxindole (3),以及16個已知化合物。從Xylaria papulis中獲得兩個新化合物:xylapapuside A (21)與xylapapuside B (22)以及五個已知化合物。在Mycoleptodiscus sp. 09F0149中分離到七個已知化合物。於Lachnum abnorme中分離到兩個新化合物:lachbenzooxocinol (30)及lachabnormic acid (31),以及八個已知化合物。 活性實驗中,hypoillexidiol (2)及(S)-(–)-3-hydroxy-3-methyloxindole (3)具有良好的抑制NO產生之活性,IC50值分別為17.5±1.8及24.7±1.6 μM,且優於正向對照組quercetin (35.9±1.7 μM);hypoillexidiol (2)、(S)-(+)-vermelone (4)、(3R)-5- methylmellein (5)及tyrosol (13)具有抑制IL-6產生之活性,IC50值為27.2±1.8~35.5±5.8 μM.
Hypoxylon sp. 12F0687 (Xylariaceae), Xylaria papulis (Xylariaceae), Mycoleptodiscus sp. 09F0149 (Magnaporthaceae), and Lachnum abnorme (Hyaloscyphaceae) are endophytic fungi, isolated from Taiwanese plants, Ilex formosana, Lepidagathis stenophylla, Piper sintenese, and Ardisia cornudentata, respectively. Investigation of these fungi led to the isolation of seven new compounds, with a compound previously unknown in nature, alone with 29 known compounds. The structures of these compounds were elucidated by spectroscopic methods. Hypoxylon sp. 12F0687 led to the isolation of three new compounds, hypoxyphenone (1), hypoillexidiol (2), and (S)-(+)-vermelone (4), with (S)-(–)-3-hydroxy-3-methyloxindole (3), previously unknown in nature, alone with 16 known compounds. From Xylaria papulis, two new compounds, xylapapusides A and B (21 and 22), together with five known compounds were isolated. Mycoleptodiscus sp. 09F0149 led to the isolation of seven known compounds. Two new compounds, lachbenzooxocinol (30) and lachabnormic acid (31), alone with eight known compounds were isolated from Lachnum abnorme. Of these isolates, hypoillexidiol (2) and (S)-(–)-3-hydroxy-3-methyloxindole (3) showed nitric oxide (NO) inhibitory activity with IC50 values of 17.5±1.8 and 24.7±1.6 μM, respectively, stronger than the positive control, quercetin (35.9±1.7 μM). Hypoillexidiol (2), (S)-(+)-vermelone (4), (3R)-5-methylmellein (5), and tyrosol (13) showed moderate inhibition of interleukin-6 (IL-6) production, with IC50 values ranging from 27.2±1.8 to 35.5±5.8 μM.
