牛樟芝苯類及三萜類成分品質管制之開發及應用

摘要


牛樟芝,一種台灣特有及珍貴的菇菌,廣泛地使用於亞洲的民俗療法上,常被用來治療癌症、肝炎及高血壓。牛樟芝子實體生長的速度極為緩慢,且一年增長2.5 平方公分的大小,導致其產品的昂貴。因此,為了取代野生牛樟芝,不同人工栽培方式的牛樟芝子實體及菌絲體產品於近幾年被發展。儘管以人工栽培的方式成功的量產牛樟芝,但其生產及銷售仍面臨了嚴重的威脅因為缺少了可信賴的品質管制的方法來評估這些牛樟芝的產品。所以研究野生及栽培牛樟芝中主量及 微量成分含量的差異性可提供準確的影像來評估牛樟芝子實體及菌絲體栽培產品的品質及其生物活性。 野生牛樟芝的成分中,苯類及三萜類化合物不僅是主量及微量的成分,亦被視為抗發炎及抗癌的活性成分。本研究中,我們開發了一種新方法以核磁共振光譜儀、高效能液相層析儀、氣相層析串連三段式四級棒質譜儀及高效能液相層析串連三段式四級棒質譜儀進行牛樟芝主量苯類及三萜類化合物的定性、半定量和定量分析。此研究以苯類及三萜類化合物為化學及生物活性的指標,提供有效品質管制的平台來評估野生牛樟芝及其相關產品。

Antrodia cinnamomea (AC), an endemic and rare Taiwanese mushroom, is widely used in Asian folk medicine for the treatment of cancer, hepatitis and hypertension. AC fruiting bodies grow in an extremely slow rate as they need one year to grow to the size of 2.5 square centimeters resulting in expensive marketed products. Therefore, different artificial cultivation of AC fruiting bodies and mycelia products are developed to replace wild AC products in recent years. Despite the success in mass production of AC by artificial cultivation, its production and marketing face a critical threat due to lack of the reliable quality assessment methods to evaluate AC-based products. Accordingly, studying concentrations in major and minor components in wild and cultivated AC will provide a clear picture for quality and biological activity of cultivated products of fruiting bodies and mycelia. Among wild AC components, benzenoids and triterpenoids not only are minor and major compounds but also play an important role for anti-inflammatory and anticancer activities. In the study, we developed a new approach for characterizing, semi-quantifying and quantifying the major AC benzenoids and triterpenoids by methods of nuclear magnetic resonance spectrometry (NMR), high performance liquid chromatography (HPLC), gas chromatography coupled to triple quadrupole mass spectrometry (GC-tandem MS), and liquid chromatography coupled to triple quadrupole mass spectrometry (HPLC-tandem MS). The present investigation will provide an efficient quality control platform to evaluate wild AC and its related products utilizing benzenoids and triterpenoids as chemical and bioactive markers.
