南仁五月茶 (Antidesma hiiranense Hayata)根部之乙酸乙酯及水可溶部進行三種癌細胞株(MCF-7, NCI-H460與SF-268)細胞毒測試,確認活性層為乙酸乙酯層,此活性層進行矽膠管柱層析,得13個分畫,其中第4~7分畫具有細胞毒活性。將活性分畫利用層析法分離,共分得27個化合物,其中3個新化合物為,barbatumol C (8), barbatumol D (9), antidesnenol (15)已及已知化合物24個為braylin (1), scopoletin (2), cedrelopsin (3), barbatumol A (4), barbatumol B (5), 3-(1,1-Dimethylallyl)scopoletin (6), obtusidin (7), antidesmanin A (10), antidesmanin B (11), antidesmanin C (12), antidesnone (13), antidesnol (14), vanillin (16), vanillic acid (17), 4-hydroxybenzaldehyde (18), syringaldehyde (19), 4-hydroxybenzoic acid (20), squalene (21), betulinic acid (22), syrungaresinol (23), ??-tocopherol (24), ??-tocopherol quinone (25), β-sitosterol (26)與 stigmasterol (27)混合物。目前材料所分離得化合物之細胞毒篩選已送測中。
Antidesma hiiranense Hayata (Euphorbiaceae) is an endemic shrub distributed throughout in the southern and eastern parts at low altitudes in Taiwan. The methanolic extract of the root of this species has shown cytotoxicity on high-throughput screening against MCF-7, NCI-H460 and SF-268 cancer cell lines, and was partitioned into EtOAc-, H2O-soluble layers and residue. Furthermore, the EtOAc-soluble layer showed cytotoxicity against above three cancer cell lines. Investigation of the active EtOAc-soluble layer by repeated chromatography and prep. TLC has led to the isolation of 3 new compounds: barbatumol C (8), barbatumol D (9), antidesnenol (15), along with 24 known compouns: braylin (1), scopoletin (2), cedrelopsin (3), barbatumol A (4), barbatumol B (5), 3-(1,1-dimethylallyl)scopoletin (6), obtusidin (7), antidesmanin A (10), antidesmanin B (11), antidesmanin C (12), antidesnone (13), antidesnol (14), vanillin (16), vanillic acid (17), 4-hydroxybenzaldehyde (18), syringaldehyde (19), 4-hydroxybenzoic acid (20), squalene (21), betulinic acid (22), syrungaresinol (23), ??-tocopherol (24), ??-tocopherol quinine (25), ??-sitosterol (26), stigmasterol (27). The structures of these compounds were elucidated by spectral analysis. The bioactivity assay against the MCF-7, NCI-H460 and SF-268 cancer cell lines are investigated in progress.
