風茹草在正常及糖尿病小鼠降血糖作用之研究

摘要


糖尿病是現今聞名的代謝性疾病,臨床上發現代謝症候群相關疾病有逐年增加的趨勢,相對代表在治療糖尿病藥物開發新藥的需求增加。風茹草是民間用的草藥。風茹草已被證實具有抗氧化和抗發炎,本論文研究目的是實驗風茹草在糖尿病鼠降血糖的作用機制。 本研究中,利用靜脈注射鏈脲佐菌素(STZ) 75 mg/kg 和給予高油高糖飲食飼養四週,誘發糖尿病小鼠動物模式。分別口服給予風茹草不同劑量1 mg/ml、5 mg/ml和10 mg/ml,對於糖尿病鼠降血糖作用和胰島素釋放的變化。在C2C12小鼠肌母細胞,實驗風茹草對於葡萄糖轉運第四型蛋白質(Glut4)表現量的影響。 實驗結果顯示,口服給予風茹草90分鐘後,發現在正常及糖尿病小鼠可以產生劑量相關性的降血糖作用。小鼠給予風茹草治療四週,發現可以改善糖尿病小鼠骨骼肌中Glut4蛋白質的表現量。在C2C12小鼠肌母細胞中,分別給予風茹草不同劑量0.01 mg/ml、0.05 mg/ml和0.1 mg/ml,反應60分鐘後,發現風茹草產生劑量相關性活化Glut4蛋白質的表現量,以0.1 mg/ml的劑量增加表現最為顯著。 實驗結果發現,風茹草確實具有調節血糖的作用,長期治療糖尿病鼠可以活化Glut4蛋白質的表現量。因此,風茹草用於治療糖尿病作為降血糖新藥開發極具潛力。

All over the world the diabetes mellitus (DM) is one of the most common metabolic disease of people’s health. Therapeutic medicine of diabetes development the increase that demand stands opposite each other at in new medicine. Glossogyne tenuifolia (GT) is a traditional Chinese herb. GT has been shown to possess antioxidative and anti-inflammatory. The aim of this research is to experiment GT relevance to antihyperglycemic. In this study, we used streptozotocin (STZ) 75 mg/kg intravenous (i.v.) injections in the lateral tail vein and high fat diet (HFD) fed mice for 4 weeks induced diabetic mouse models. The effects of GT (p.o.) on animals showing blood glucose levels and release insulin. In C2C12 myoblasts, the effects of GT on glucose uptake, activity of glucose transporter type 4 (Glut4) and glycogen synthesis were examined. The experimental result of this study is found in acute treatment with GT (p.o.) produced a hypoglycemic effect in a dose-dependent manner in normal and diabetic mice, GT significantly lowered the plasma glucose at 90 min after oral treatment. Four-week treatment of diabetic mice with GT improved the expression of the Glut4 in the skeletal muscles. In C2C12 myoblasts were treated GT with different concentrations of 0.01 mg/ml, 0.05 mg/ml and 0.1 mg/ml in 60 minutes. Treating with GT resulted increased of Glut4 expression in C2C12 myoblasts concentration-depende- ntly, Glut4 expression was significantly increased at 0.1 mg/ml. Found by the results, GT does have the effect of the regulation of blood glucose. Hence, GT have potential as an antidiabetes medicine for the treatment of DM.
