利用中孔洞二氧化矽氣凝膠吸附Benzophenone-3提升防曬效能之研究

摘要


本研究利用室溫離子熔液作為模板,以溶膠凝膠法進行反應,製備具有高表面積之中孔洞二氧化矽氣凝膠(MS)作為載體,吸附有機防曬劑BP-3,製成MS吸附BP-3 之複合型防曬劑,AS5B1及AS4B1,並進行測試與分析。由TGA、DSC及UV-VIS結果得知,MS吸附BP-3之含量分別約為34wt% (AS5B1)及45 wt% (AS4B1),因MS之吸附大幅降低BP-3在這兩個複合型防曬劑的結晶性,並且呈現協同效應,提高紫外線吸收效果。而UVB及UVA之防曬係數(SPF及UVA)、防曬係數穩定性與光穩定性測試結果顯示,含AS5B1防曬乳液(AS5B1-E)的SPF與UVA值比添加等量BP-3(2 %)之值分別提升17.3 % 和17 %,且MS吸附BP-3後有效的降低防曬係數隨時間降低之速度,並明顯降低紫外光對防曬劑的破壞,證實MS對BP-3之吸附可有效提高其對紫外線之防護效果、防曬效果的穩定性,及防曬劑之光穩定性。此外,體外釋放及經皮滲透測試結果進一步發現,當MS吸附BP-3後,有效降低BP-3在三酸甘油酯中的釋放,而BP-3之經皮滲透亦有效降低,將可有效減少對皮膚之傷害。 以上結果顯示,MS作為有機防曬劑之載體,可提高防曬化妝品的有效性及安全性,未來應用在防曬劑與其他外用藥物上,具有潛在的發展性。

In this study, various amounts of benzophenone-3 (BP-3) were adsorbed in a high surface area mesoporous silica (MS), as a drug carrier, which was synthesized via the sol-gel reaction with a room temperature ionic liquid as the pore formation template, to form the MS-carried BP-3 sunsceens, AS4B1 and AS5B1. The as-prepared sunsceens were characterized and analyzed by TGA, DSC and UV-VIS measurements. The results showed that the amount of BP-3 loaded in MS was about 45 wt % and 34 wt % for AS4B1 and AS5B1, respectively, the MS carrier adsorption effectively reduced the BP-3’s crystallinity and a synergistic UV protection effect was found in the as-prepared sunscreens. Besides, the stability of the UV protectoon ability and the photostability of the sunscreen agents, as well as the in vitro sun protection factors of UVB and UVA for the AS5B1-based sunscreen were also investigated. The results indicated that because BP-3 was carried by the MS matrix, the in-vitro SPF and UVA value of the AS5B1-based sunscreen was about 17.3 % and 17.0 % higher than that of the free BP-3-based sunscreen, respectively, the sun protection factors were reduced more slowly with increasing time, and the the photodegradation of the screen was significantly lowered. These revealed that after adsorption by MS, the UV protection ability and protection stability of the AS5B1-based sunscreens in both the UVB and UVA domains as well as the photostability of the UV screens were all significantly enhanced. Further, the results of release of BP-3 in triglyceride and in vitro permeation test revealed that the release of BP-3 from AS5B1 and its percutaneous penetration was largely decreased after adsorption by MS, avoiding a direct contact with skin and reducint the harm to skin. This study introduced the as-prepared MS as a carrier for BP-3 to increase efficacy and safety of the sunscreen, revealing that the adsorption by MS is a good technological strategy for the sunscreen cosmetics and MS is a promising organic sunscreen drug carrier and should be a potential carrier for other topical drugs as well.
