摘要


目前生物科技中,可以藉由不同層面的方式最佳化(optimize)異 源蛋白表現量(heterologous protein expression),包括調整基因 或是培養細胞條件等。而在基因層面中,使用密碼子(codon)做為調 整的依據是做法。在眾多蛋白質表現系統中如大腸桿菌、酵母菌、哺 乳類等,由於其基因組的定序已完成,因此外源蛋白的目標基因可以 依據所表現的細胞的基因來修正。而昆蟲的桿狀病毒表現系統由於其 宿主鱗翅目(Lepidoptera)家蠶(Bombyx mori)的基因組在 2008 年才 完成定序,因此目前的文獻對於在桿狀病毒表現系統中,調整密碼子 來增加蛋白質產量的研究不多。我們藉由研究桿狀病毒以及家蠶的基 因組,希望能找到最佳化的方式。然而在之前文獻中指出,單純將外 源基因的密碼子替換成宿主的最佳密碼子,並不保證能增加蛋白質產 量。還需考慮基因中的 GC 比、mRNA 穩性性、mRNA 的二級結構、特殊 序列、密碼子對、限制酶切割位、tRNA 含量等。研究中,分成三大部 分:第一部分為計算及觀察三種桿狀病毒基因組、鱗翅目細胞基因組。 第二部分將比較這些基因特性跟現有的 mRNA 和蛋白質表現量並找出 其中關連性。第三部分將參考上述資料,設計適用於昆蟲桿狀病毒表 現系統中,調整基因以改善蛋白質表現的方法。

In current biotechnology, the heterologous protein expression can be optimized in various level, including adjustment to the gene (codon) or cell culture co_nditions. In many protein expression systems such as E. coli,yeast, mammalian, etc., because of its genome sequencing has been completed, so the heterologous gene can be modified. Because genome of silkworm (Bombyx mori) was completed sequences in 2008, the current research about optimization gene expression in the baculovirus expression system is absent. By studying the genome of the silkworm and baculovirus,we hope to find the best optimization method. However, the study indicates that simply replacing the codons of the exogenous gene into the host optimal codons are not guaranteed to increase protein yield. We also need to consider the gene GC ratio, mRNA stability of the secondary structure of mRNA, special sequences, codon pairs, restriction enzyme cleavage, tRNA content and so on. Three parts in this study: the first part is calculation and observation codons and other features in three kinds of baculovirus genome and lepidopteran insect cell genome. The second part we will compare these characteristics with mRNA expression and to identify which relation. The third part we use above information to adjust the codon usage for improvement of protein expression.
