白藜蘆醇及其衍生物對於抑制發炎反應與抗自由基作用之探討

Effects of Anti-inflammation and Anti-free Radicals of Resveratrol and Its Derivatives

摘要


白蔾蘆醇(Resveratrol 3, 5, 4-trihydroxystilbene) 為存在植物中之多酚類產物,為植物之防禦素。由過去文獻了解到白蔾蘆醇及其衍生物具有抗氧化、抗發炎及抗癌等能力。最近的研究也顯示白蔾蘆醇有增加老鼠、果蠅及魚類壽命、抑制肝腫大、糖尿病及心血管發炎的效益。 本實驗將白蔾蘆醇氫氧基與硫、氮離子做替代交換並純化後所得衍生物做為測試藥物並以 macrophage cell line RAW264.7 為實驗對象。實驗目的分兩部份。第一部分,利用DPPH 觀察白蔾蘆醇及其衍生物清除自由基的能力,接著使用細胞內毒素(LPS)誘使RAW264.7細胞發炎以研究白蔾蘆醇及其衍生物對於細胞的保護作用與抑制發炎反應的效果。第二部份,我們用H2O2 誘使細胞產生脂質過氧化現象,並利用MDA assay 來觀察白蔾蘆醇及其衍生物對於抗自由基所產生的效果。 結果顯示,編號從AnWen-1031到 AnWen-1050的測試藥物中,有六種較具清除自由基能力(AnWen-1031、1033、1034、1035、1036、1038)。而這六種藥物對於細胞內毒素(LPS)所誘導NO的產生均有抑制效果。在保護細胞避免脂質過氧化傷害效果方面,經過氧化氫200μM濃度刺激,AnWen-1031、1033、1034、1038對細胞存活率沒有提升效果,AnWen-1035、1036、卻可增加細胞存活率。而在MDA assay方面,白蔾蘆醇等六種測試藥物均有降低因過氧化氫(H2O2)刺激所產生脂質過氧化傷害能力。 綜合上述結果,白蔾蘆醇及其衍生物在清除自由基、抗發炎及保護細胞脂質過氧化傷害作用方面,在特定濃度範圍下有一定程度的效果。因此,在未來作為藥劑的發展潛力上可做更深入的研究探討。

Resveratrol 3, 5, 4-trihydroxystilbene is a polyphenolic compound that act as a phytoalexin in plants. In according to past references, resveratrol and its derivatives have the ability of anti-oxidation, anti-inflammation and anticancer. Resveratrol can increase life span of rats, fruit fly and rabbits. It also has the effectiveness of inhibition of liver tumor, diabetes and cardiovascular inflammation. We used the macrophage cell line RAW264.7 in the study. Hydrogen was replaced by sulfer and nitrogen of the reveratrol. And then, we purified its derivatives to act as drugs. The study was proceeded in two phases. First, we used DPPH to observe the ability of resveratrol and its derivatives in eliminating the free radicals and then studied the protectiveness and anti-inflammation of resveratrol and derivatives when stimulated RAW264.7 cell line by lipopolysaccharide. Second, we used MDA assay to observe the protective effective of resveratrol and derivatives interacting with the free radicals. The AnWen derivatives from 1031 to 1050, there were six derivatives which had the ability of elimination of free radicals (1031, 1033, 1034, 1035, 1036 and 1038). Also, these six derivatives had the inhibitive effect in NO production stimulated by lipopolysaccharide. AnWen 1031, 1033, 1034 and 1035 had no upregulative effect stimulated by hydrogen peroxide with regard to cell viability. However the AnWen 1036 and 1038 could increase the cell viability stimulated by hydrogen peroxide. Resveratrol and its six derivatives all decreasef the damage stimulated by hydrogen peroxide. In short, Resveratrol and its six derivatives had the effects of anti-free radical, anti-inflammation and protection in lipid peroxidation.
