中草藥九節茶應用於尋常性痤瘡之修復

Application of herbal sarcandra in remediation of acne vulgaris

摘要


九節茶(Sarcandraglabra)別名草珊瑚、觀音茶、接骨木,為長年生常綠灌木,植物中含九節風內脂A及D(chlorantha lactoneA及D)、鞣質(tannins)、琥珀酸(IUPAC)等成份及防禦素。先前發表的文獻記載九節茶具有增強免疫功能、祛風除濕及活血止痛的功效。近期的研究也顯示了九節茶萃取物除了具有促進傷口癒合的能力外,亦能改善小鼠體內腫瘤細胞的生長繁殖能力,提高過氧化氫酶(Catalase)的活力,對癌細胞的耗氧能力有直接抑制作用,改善能量代謝而實現抗癌作用。本研究則是採用中草藥植物九節茶作為尋常性痤瘡的修復成分,並深入檢測藥材對痤瘡形成因素的改善成效。 本實驗所用九節茶藥材是以其根莖部位萃取使用,實驗以五階段逐次進行。第一階段為將中草藥植物碎化後以乙醇泡製,再經由索氏(Soxhlet)萃取成實驗藥材。第二階段為細菌實驗,針對痤瘡丙酸桿菌(Propionibacterium acnes)及金黃色葡萄球菌(Staphyloccocus aureus)二種引發及存在於痤瘡患部促發炎症的細菌,實驗其抑菌功效。第三階段是利用RAW264.7巨噬細胞(Macrophage)檢測細胞存活率(MTT Assay)。第四階段為測試藥材對引發炎症的毒性物質防禦效果。第五階段為DPPH自由基活性清除率之檢測。由此五個階段的實驗,探討中草藥植物九節茶萃取物應用於修復尋常性痤瘡的可行性。 實驗結果顯示中草藥植物九節茶萃取物在痤瘡丙酸桿菌和金黃色葡萄球菌抑菌實驗中均能產生有效的抑菌範圍,發揮抑菌效果。再將藥材以逐次對半稀釋的方式採5組測試組(0.5%、0.25%、0.125%、0.0625%、 0.03125%)和控制組分別做細胞存活率、抗發炎及抗氧化能力實驗,其結果皆有明顯的提升。MTT assay細胞活化能力及DPPH自由基活性清除皆提升了3倍的有效率;細胞抗發炎實驗中加入含毒性因子的LPS,讓細胞達到完全發炎的狀態,經施於定量藥材後,約能達到90%之抵禦毒性物質的傷害,有效發揮保護細胞的機制。經由以上五個階段的實驗,顯示九節茶萃取物具備了研發成為治療尋常性痤瘡的條件。 中草藥保養品的研發一直是廣受注目的,尤其是針對尋常性痤瘡的治療;痤瘡患部若長期以化學藥物治療(如:抗生素)會產生具有抗藥性的細菌而造成日後醫療上的難題;而溫和的中草藥成分產生抗藥性的顧忌極低,因此本研究結果可作為修復尋常性痤瘡產品研發的參考依據。

Sarcandra glabra also known as grass coral, guanyin tea or elderberry is an evergreen shrub plant containing clorantha lactone A and D, tannins, succinate, etc. Published literatures have shown that Sarcandra glabra with enhanced immune function and efficacy can get rid of rheumatism. Recent studies have also reported that Sarcandra glabra have the ability to promote healing and inhibit the growth and reproduction ability of tumor cells in mice. Researches have shown that the herb can increase catalase enzyme activity of cancer cells in mice and improve tumor-bearing capacity of the body’s oxygen consumption which has a direct inhibitory effect on cancer cells. This study was to use herbal plants sarcandra repair component as acne vulgaris, and further improve the effectiveness of herbs to detect the formation of acne factors. This feasibility study presented five stages of experiments to explore the ability of Sarcandra glabra herbal exctract as a repair component in acne vulgaris. The drug’s inhibitory effect, cell viability, anti-inflammatory and anti-oxidative response capability (free radicals) in the treatment of acne vulgaris were assessed. The sample was prepared by shredding the rhizomal parts of the herbal plant using ethanol followed by brewing. The test drug was extracted using ultrasound-assisted Soxhlet extraction.Antibacterial efficacy experiments for Propionibacterium acne and Staphylococcus aureus Rosenbach were performed and RAW264.7 macrophage cell line was used for cell viability test (MTT assay). The inhibitory effect of the test drug for NO release was also studied using Nitric Oxide Assay followed by anti-free radical DPPH test. Where by the five stages of the experiment, the feasibility of herbal plant extracts applied sarcandra repair acne vulgaris. Experimental results showed that sarcandra glabra extracts was able to effectively inhibit bacterial activities of propionibacterium acne and staphylococcus aureus. Comparison of the increasing dilution in the five test groups(0.5%,0.25%,0.125%,0.0625%,0.03125%) and control group in terms of cell viability, anti-inflammatory and antioxidative capacity showed significant improvements.MTT assay cell activation ability and DPPH radical scavenging activity are to enhance the efficiency of 3 times.Anti-inflammatory cell experiments induced by LPS showed inflammatory response, but after the addition of herbs, which can achieve above 90% against of the resistance to toxic substances and play a defense mechanism to protect the cells. Chinese herbal skin care products research and development has been a high-profile especially for the treatment of acne vulgaris. Long-term treatment of acne with chemical drugs(eg:antibiotics) will produce drug-resistant bacteria and cause medical problems in the future and mild herbal ingredients resistant scruples low, therefore the results of this study can be used as a product development reference in the treatment of acne vulgaris.
